基于HPLC指纹图谱和数学判别模式的丹参道地性研究

摘　　要：目的:建立基于数学判别模式和HPLC指纹图谱方法应用于丹参道地性判别。方法:采用Symmetry C18色谱柱(250mm×4.6 mm,5μm),以0.1%磷酸水-乙腈为流动相进行梯度洗脱,流速为1.0 ml·min^-1,检测波长为270 nm,柱温为25℃,运用"中药色谱指纹图谱相似度评价系统(2012 A版)"分别对10批四川中江道地丹参与8批非道地丹参进行相似度评价,运用SPSS 22.0对不同指纹图谱进行聚类分析与判别分析,为丹参道地性的分类提供理论依据。结果:建立的HPLC指纹图谱精密度、重复性和稳定性均良好,采用HPLC指纹图谱和判别分析相结合的方法可以得到四川中江丹参的道地性判别函数,回代验证准确率达到100%(n=18),显示该方法可行、有效。结论:丹参HPLC指纹图谱结合判别分析方法是一种科学有效的判别丹参药材道地性的方法。Objective:To establish an HPLC fingerprint for the genuineness discrimination of Salvia miltiorrhiza Bge..Methods:The HPLC separation was performed on a Symmetry C18(250 mm×4.6 mm,5μm)column with the mobile phase consisting of water(0.1%phosphoric acid)-acetonitrile with gradient elution at a flow rate of 1.0 ml·min^-1.The detection wavelength was set at 270nm and the column temperature was 25℃.The HPLC fingerprints of samples from ten genuine and eight no-genuine producing areas of Salvia miltiorrhiza Bge.were created to access the similarities,and the fingerprints were identified by SPSS 22.0 software for cluster analysis and pattern discrimination mode.Results:The established fingerprint of Salvia miltiorrhiza Bge.was specific,precise and stable.Discrimination function in the genuineness discrimination for Salvia miltiorrhiza Bge was useful and effective with the accuracy of regeneration verification of 100%(n=18).Conclusion:The numerical pattern discrimination combined with HPLC fingerprint can be used in the genuineness discrimination of Salvia miltiorrhiza Bge effectively.
