摘  要:目的观察曲克芦丁预处理对大鼠缺血再灌注心肌的保护作用。方法健康雄性Wistar大鼠24只,随机分为假手术组(Sham组)、缺血再灌注组(I/R组)和曲克芦丁预处理组(I/R+TRX组)。I/R+TRX组在建模前4周进行曲克芦丁(150mg/kg体重)口服给药,Sham组和I/R组口服生理盐水。结扎左冠状动脉前降支的方法建立大鼠心肌缺血再灌注损伤模型。检测各组大鼠心功能及血流动力学变化,氯化三苯基四氮唑(TTC)染色检测大鼠心肌梗死面积,原位缺口末端标记法(TUNEL)染色检测大鼠心肌细胞凋亡,放射免疫法测定肌钙蛋白I(cTnI)、肌酸激酶(CK)、乳酸脱氢酶(LDH)水平,酶联免疫吸附试验(ELISA)法测定肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-10含量。应用SPSS19.0统计软件分析。数值变量采用均值±标准差(Mean±SD)表示,两组之间比较采用t检验,多组之间比较采用单因素方差分析。结果与缺血再灌注组比较,曲克芦丁预处理组大鼠心肌梗死面积明显减小,差异有统计学意义(P<0.05)。与假手术组比较,缺血再灌注组大鼠血液动力学变化差异有统计学意义(P<0.05);与缺血再灌注组比较,曲克芦丁预处理组大鼠血液动力学变化差异有统计学意义(P<0.05);与假手术组比较,曲克芦丁预处理组大鼠血液动力学变化差异无统计学意义(P>0.05)。与假手术组比较,缺血再灌注组大鼠血清cTnI、CK、LDH、TNF-α、IL-10变化差异均有统计学意义(P<0.05);与缺血再灌注组比较,曲克芦丁预处理组大鼠血清cTnI、CK、LDH、TNF-α、IL-10变化差异均有统计学意义(P<0.05);与假手术组比较,曲克芦丁预处理组大鼠血清cTnI、CK、LDH、TNF-α、IL-10变化差异均有统计学意义(P<0.05)。心肌细胞凋亡率假手术组为(2.87±0.32)%,缺血再灌注组为(31.25±2.76)%,曲克芦丁预处理组为(18.43±2.53)%。与假手术组比较,缺血再灌注组大鼠心肌细胞凋亡率差异有统计学意义(P<0.05);与�Objective To investigate the protective effects of troxerutin preconditioning on ischemia-reperfusion myocardial in rats. Methods 24 male Wistar rats were divided into sham group, ischemia/reperfusion group (I/R group) and troxerutin preconditioning group (I/R+ TRX group) randomly. The myocardial ischemia-reperfusion injury rat model was created with troxerutin preconditioning. Myocardial infarct size was evaluated by the Evans blue-triphenyltetrazolium chloride (TTC) method. Cell apoptosis was detected by terminal-deoxynucleotidyl transferase mediated nick end labeling (TUNEL) method. Hemodynamic parameters and cardiac function were monitored. Serum cardiac troponin I (cTnI), creatine kinase (CK), lactate dehydrogenase (LDH), tumor necrosis factor-α(TNF-α) and interleukin (IL)-10 were measured. Results Myocardial infarct size in I/R+ TRX group was significantly decreased than that in the I/R group (P<0.05). There were significant difference in Hemodynamic parameters and cardiac function between sham group and I/R group (P<0.05);There were significant difference in Hemodynamic parameters and cardiac function between I/R group and I/R+ TRX group (P<0.05);There were no significant difference in Hemodynamic parameters and cardiac function between sham group and I/R+ TRX group (P>0.05). Serum cTnI, CK, LDH, TNF-α and IL-10 concentration in I/R group were significantly higher than those in sham group and I/R+ TRX group (P<0.05);Serum cTnI, CK, LDH, TNF-α and IL-10 concentration in I/R+ TRX group were significantly higher than those in sham group (P<0.05). Myocardial apoptosis rate were (2.87±0.32)% in sham group,(31.25±2.76)% in I/R group and (18.43±2.53)% in I/R+ TRX group respectively. Myocardial apoptosis rate in I/R group was significantly higher than that in sham group and I/R+ TRX group (P<0.05);There was no significant difference in myocardial apoptosis rate between sham group and I/R+ TRX group (P>0.05).
