蟠桃丸化裁方对自发性高血压大鼠主要安全性指标的影响

摘　　要：目的探讨常规使用蟠桃丸化裁方对自发性高血压大鼠(SHR)主要安全性指标的影响。方法以Wistar kyoto(WKY)大鼠为对照组,SHR随机均分为模型组、厄贝沙坦+多奈哌齐组、多奈哌齐组、蟠桃丸化裁方高剂量组和蟠桃丸化裁方低剂量组,每组12只,雌雄各半,给药11周后检测红细胞、血红蛋白、胆红素、肝肾功能指标、血清胆碱酯酶等指标。结果各组间红细胞计数、红细胞比容、红细胞平均体积、红细胞分布宽度等溶血相关指标比较差异无统计学意义( P >0.05)。与SHR模型组比较,蟠桃丸化裁方低剂量组平均血红蛋白浓度、间接胆红素、直接胆红素显著降低( P <0.05或 P <0.01),高剂量组血红蛋白、平均血红蛋白浓度均显著降低( P <0.05或 P <0.01);西药组胆红素、血红蛋白等指标差异无统计学意义( P >0.05)。与SHR模型组相比,多奈哌齐组、蟠桃丸化裁方高剂量组与低剂量组血清胆碱酯酶水平明显升高( P <0.01);厄贝沙坦+多奈哌齐组血清胆碱酯酶水平差异无统计学意义( P >0.05)。模型组天门冬氨酸氨基移酶(AST)、γ-谷氨酰转移酶(γ-GT)、尿酸等肝肾功能指标较对照组明显升高( P <0.05或 P <0.01),而蟠桃丸化裁方组ALT、AST、γ-GT、尿素、尿素氮、肌酐、尿酸与模型组比较差异均无统计学意义( P >0.05),且肌酐、尿酸水平有下降趋势。结论常规使用蟠桃丸化裁方对SHR无明显肝肾损伤,未见明显溶血反应,且能上调血清胆碱酯酶水平,因此蟠桃丸化裁方具有一定的安全性。Objective To investigate the effects of conventional use of the cutting formula of Pantao Pills on the main safty indicators in spontaneously hypertensive rats (SHR). Methods Wistar kyoto (WKY) rats were used as control group.SHR were randomly divided into model group,irbesartan+donepezil group,donepezil group,low-dose group of the cutting formula of Pantao pills,and high-dose group of the cutting formula of Pantao pills.Twelve rats in each group were tested for red blood cells,hemoglobin,bilirubin,liver and kidney function indicators,cholinesterase,and other indicators after 11 weeks of administration. Results There were no significant differences in hemolysis-related indicators between groups,including red blood cell count,hematocrit,mean red blood cell volume,and red blood cell distribution width ( P >0.05).Compared with the SHR model group,the average hemoglobin concentration,indirect bilirubin,and direct bilirubin in the low-dose group of the cutting formula of Pantao pills were significantly lower ( P <0.05 or P <0.01),and the hemoglobin and mean hemoglobin concentrations in the high-dose group was significantly decreased ( P <0.05 or P <0.01).There were no significant differences in the indicators of bilirubin and hemoglobin in the western medicine group( P >0.05).Compared with the SHR model group,serum cholinesterase levels were significantly increased in the donepezil group,high-dose of the cutting formula of Pantao pills,and low-dose groups ( P <0.01),There was no significant difference in serum cholinesterase in the irbesartan+donepezil group ( P >0.05).The liver and kidney function indexes of the model group such as AST,γ-GT,and uric acid were significantly higher than those of the control group( P <0.05 or P <0.01),There was no significant difference in ALT,AST,γ-GT,urea ,and urea nitrogen between the cutting formula of Pantao pills group and the model group( P >0.05),and the levels of creatinine and uric acid decreased in the cutting formula of Pantao pills group.
