摘  要:为了创制一批携带优良农艺性状的番茄种质资源新材料,以栽培番茄品种'Jina'(Solanum lycopersicum)为受体亲本,以野生醋栗番茄LA2093(S.pimpinellifolium)为供体亲本,利用杂交、3次回交和InDel、SSR分子标记辅助选择方法构建了一套以栽培番茄'Jina'为遗传背景、含LA2093的基因的渐渗系。从BC1群体构建的遗传图谱中选择均匀分布于12条染色体的96对InDel、SSR分子标记追踪野生番茄的渗入片段,在BC3代共留选渐渗系材料51份,其中单片段材料14份、双片段材料27份、三片段材料10份;共得到渗入片段98个,平均单个渗入片段长度为35.69cM;51份渐渗系平均渗入供体基因片段占受体基因组的比例为5.6%,完全覆盖12条染色体。利用BC3F1群体检测到与首花序节位、花序间隔节位、单花序花数3个农艺性状相关的6个QTL位点,单个位点的贡献率为4.33%~47.93%。In order to create a batch of new tomato germplasm resources with excellent genes, in this study a set of introgression lines were created between the wild tomato LA2093 (the donor) and the cultivated tomato "Jina"(the receptor). Through hybridization, three-time backcrossing and InDel and SSR marker-assisted selection, a set of introgression lines have been constructed. They had the genetic background of cultivated tomato "Jina" with the fragment from LA2093. A genetic map was constructed in the BC 1 generation, and 96 pairs of InDel and SSR molecular markers uniformly distributed on the 12 chromosomes were selected to track the infiltrated fragments of wild tomato. Subsequently, 14 single-segment introgression lines, 27 double-segment introgression lines and 10 three-segment introgression lines were obtained in the BC3 generation. A total of 98 introgression segments were obtained, and the average length of a single introgression segment was 35.69 cM. Each introgression line contained an average of 5.6% of the donor gene. They overlapped and covered 100% of the wild tomato genome. In BC3F1 population, 6 QTLs were detected for 3 agronomic traits including the leaf node below the first inflorescence, the node of interval inflorescence and the number of flowers per inflorescence. The contribution rate for a single locus was 4.33%~47.93%.
