环状RNA在系统性红斑狼疮疾病中的筛选及表达

摘　　要：目的:探讨环状RNA(circRNA)在系统性红斑狼疮疾病(SLE)的表达及在SLE发病机制中的作用.方法:利用CIRCpedia数据库,筛选出候选的SLE相关环状RNA,应用荧光定量PCR(RT-qPCR)技术分析SLE患者和健康体检者血浆,筛选出目标环状RNA,再用RT-qPCR对目标环状RNA及其对应基因进行验证,最后对其进行生物信息学分析.结果:荧光定量PCR分析显示SLC15A4基因对应的环状RNA:HSA_CIRCpedia_7637在SLE患者组中明显降低,有统计学差异(P=0.0481);并验证SLC15A4基因线性RNA(P=0.0003)和环状RNAHSA_CIRCpedia_7637(P=0.0069)在SLE患者中均比健康对照者明显降低,有统计学差异.生物信息学分析表明HSA_CIRCpedia_7637比较可能结合的miRNA有11种,包括hsa-miR-638等.结论:SLE患者的环状RNAHSA_CIRCpedia_7637表达下调;HSA_CIRCpedia_7637可能通过竞争性结合hsa-miR-638来参与SLE疾病发生.Objective:To investigate the expression of circular RNA(circRNA)in systemic lupus erythematosus(SLE)and its role in pathogenesis of SLE.Methods:The candidate SLE-related circRNA was screened out using CIRCpedia database.RT-qPCR was used to detect circRNA expression in 10 patients with SLE and 10 healthy subjects.The differentially expressed target circRNA was screened out followed by validation of the target circRNA and its corresponding genes using RT-qPCR.Finally,bioinformatics analysis was carried out.Results:The expression level of HSA_CIRCpedia_7637,which corresponding to SLC15A4 gene,was significantly decreased in SLE patients,and there was a statistically significant difference(P=0.048 1).Further confirmation showed that the level of linear RNA and circular RNA(HSA_CIRCpedia_7637)of SLC15A4 gene were significantly lower in SLE patients than in healthy controls with statistical significance(P=0.000 3 and P=0.006 9).Bioinformatics analysis indicated that there were 11 kinds of miRNAs possibly bound with HSA_CIRCpedia_7637,including hsa-miR-638.Conclusion:The expression of HSA_CIRCpedia_7637 was down-regulated in SLE patients.HSA_CIRCpedia_7637 may be involved in occurrence of SLE by competitively binding with hsa-miR-638.
