摘  要:利用大孔树脂吸附作用对其中总香豆素类成分进行纯化,使用紫外分光光度计检测总香豆素的含量.通过对大孔树脂的含水量、静态和动态吸附、上柱液的体积、树脂的径高比、洗脱剂乙醇的浓度、洗脱速度、用量体积等因素对长毛风毛菊总香豆素纯化影响的研究,得出最佳纯化工艺为:以AB-8型大孔树脂,径高比为1:5,上样量2BV,75%乙醇洗脱,洗脱速度为2BV-h-1,洗脱剂用量为4BV.该纯化工艺简单高效,可为长毛风毛菊中总香豆素的资源开发和利用提供参考.To reserch the purification process of total coumarin from the extract of Saussurea hieracioides,Total coumarins was purified by macroporous resin adsorption, and the content of total coumarins was detected by UV. The effect of macroporous resin on the purification efficiency of total coumarin from S. hieracioides was evaluated by investigating the water content, static adsorption and dynamic adsorption of macroporous resin, as well as the volume of upper column liquid, the ratio of diameter to height of resin,the concentration of eluent ethanol,the elution speed of eluent ethanol,the dosage volume of eluent ethanol,etc. The optimal purification process of total coumarin from S. hieracioides was as follows: AB - 8 macroporous resin,diameter to height ratio of 1 : 5 ,sample loading amount of 2 BV, 75% ethanol elution, elution speed of 2 BV · h^-1, and dosage of remover of 4 BV. The purification process is simple and efficient,which can provide reference for the development and utilization of total coumarins in S. hieracioides.
