摘  要:目的:探讨匙羹藤总皂苷(Gymnema sylvestre total saponins,GA)对2型糖尿病(type 2diabetes mellitus,T2DM)KK-Ay小鼠胰岛素抵抗(insulin resistance,IR)的影响,并从脂肪组织Cb1/CAP信号通路介导的GLUT4转运角度探讨其作用机制。方法:将27只KK-Ay小鼠随机分为模型组(MG)、吡格列酮组(BG)、匙羹藤总皂苷组(GA),并选取9只C57BL/6J小鼠作为正常组(NG),连续灌胃给药8周,每周检测小鼠血糖水平,实验第4、8周末进行小鼠口服糖耐量试验(oral glucose tolerance test,OGTT),并计算曲线下面积AUC,给药结束后检测小鼠血清中甘油三酯(TG)、血清总胆固醇(TC)、高密度脂蛋白(HDL)、低高密度脂蛋白(LDL)含量;取小鼠附睾脂肪组织,采用实时荧光定量PCR法测定过氧化物酶体增殖物激活受体γ(PPARγ)、葡萄糖转运蛋白4(glucose transporter 4,GLUT4)、原癌基因蛋白Cb1及其接蛋白CAP的mRNA含量。结果:与NG组相比,MG组血糖升高(P<0.05)、AUC升高(P<0.01),TG、TC、LDL及HDL增加(P<0.05),PPARγ mRNA降低(P<0.01),GLUT4 mRNA降低(P<0.05)、CAP mRNA降低(P<0.01),Cb1 mRNA增加(P<0.01);与MG组相比,GA、BG组血糖降低(P<0.05),治疗第4周AUC降低(P<0.05)、第8周AUC降低(P<0.01),GA组TG及LDL降低(P<0.05);GA、BG组PPARγ mRNA增加(P<0.05)、GLUT4 mRNA增加(P<0.01)、CAP mRNA增加(P<0.01)。结论:匙羹藤总皂苷对T2DM小鼠具有降糖调脂、改善IR的作用,其作用机制可能与通过PPARγ激活Cb1/CAP信号通路,增加GLUT4转运有关。Objective: To investigate the effects of gymnema sylvestre total saponins(GA) on insulin resistance (IR) of KKAY mice with type two diabetes mellitus (T2DM), and discuss its mechanism from the angles of adipose tissue Cb1/CAP signal channel mediating GLUT4 transportation. Methods: All 27 KK-Ay mice were randomly allocated to the model group (MG), pioglitazone group(PG) and GA group, nine C57BL/6J mice were selected as the normal group (NG), and drenched for eight weeks consecutively, to detect the levels of blood glucose each week, by the end of the forth and eighth week, oral glucose tolerance test (OGTT) of the mice was performed, and the area of AUC was calculated under the curve, after medicating, to detect the contents of TG, TC, HDL and LDL;to measure the contents of peroxisone proliferator-activated receptor-γ(PPAR γ), glucose transporter 4(GLUT4), proto-oncegene protein Cb1 and mRNA content of CAP using realtime PCR after adipose tissue from epididymis of the mice was taken. Results: Compared with NG group, the level of blood glucose in MG group raised (P<0.05), AUC raised (P<0.01), the levels of TG, TC, LDL and HDL increased (P<0.05), PPAR γ mRNA lowered (P<0.01), GLUT4 mRNA decreased (P<0.05), CAP mRNA decreased (P<0.01), Cb1mRNA increased (P<0.01);compared with MG group, the levels of blood glucose in GA and PG group lowered (P<0.05), AUC decreased at the forth week of treating (P<0.05), and it decreased at the eighth week (P<0.01), TG and LDL decreased in GA group (P<0.05);PPAR γ mRNA increased in GA and PG group(P<0.05), GLUT4 mRNA increased(P<0.01), and CAP mRNA increased (P<0.01). Conclusion: Gymnema sylvestre total saponins could lower blood glucose, regulate blood lipid and improve IR of T2DM mice, and its mechanism might be related to activating Cb1/CAP signal channel and increasing GLUT4 transportation using PPARγ.
