摘 要:目的检测复合应激ED大鼠模型血清代谢组变化与伊木萨克片的影响,研究其发病机制与药物的作用机制。方法选取正常雄性大鼠150只,随机抽取30只为正常对照组(N组),余120只为造模组,通过环境雌激素样饮食联合冷刺激干预20周建立复合应激ED大鼠模型(60只),后分为复合应激ED大鼠模型组(ED组,30只)和伊木萨克片干预组(Y组,30只),干预2周后,采用1H-NMR检测各组大鼠血清代谢组变化。结果(1)ED组较N组大鼠血清中极低密度脂蛋白(VLDL)、亮氨酸、胆碱、谷氨酰胺、丙酮、缬氨酸、7-氨基丁酸、丙酮酸、谷氨酸、柠檬酸、肌酸、牛磺酸、蛋氨酸、天冬氨酸表达量降低;乳酸、丙氨酸、乙酰乙酸、氧化三甲基胺、P-葡萄糖、胍基乙酸、呋喃糖表达量升高,差异有统计学意义(P<0.05);(2)Y组较ED组大鼠血清中乳酸、丙氨酸、氧化三甲基铵,P-葡萄糖、胍基乙酸、P-呋喃糖表达量降低,VLDL和胆碱表达量增高,差异有统计学意义(P<0.05)。结论复合应激ED大鼠模型出现能量和三大物质代谢紊乱,而伊木萨克片干预后可能通过纠正代谢紊乱进而抑制炎症、氧化应激和内皮功能障碍等机制,促进ED的恢复。Objective To analyze the changes in serum metabolomics of compound stress ED rat models and their medicine intervention, and explore its pathogenesis and intervention mechanism. Methods Total of 150 normal mature male rats were selected in this study, 30 of them were randomly set as the normal group (N group), the remaining 120 rats were as the model group, the compound stress ED rat model (60 rats) was established by environmental estrogen-like diet combined with cold stress intervention for 20 weeks, then screened into compound stress ED model group (ED group, 30) and Yimusake tablet intervention group (Y group, 30). After 2 weeks of intervention, the changes of serum metabolomics in each group were detected by 1H-NMR. Results (1) The serum levels of VLDL, leucine, choline, glutamine, acetone, valine, gamma-aminobutyric acid, pyruvate, glutamic acid, citric acid, creatine, taurine, methionine and aspartic acid in ED group were lower than those in N group, while the levels of lactic acid, alanine, acetoacetic acid, trimethylamine oxide, beta-glucose, guanylacetic acid and beta-furose in ED group were higher than those in N group (P< 0.05).(2) The expression of lactic acid, alanine, trimethylammonium oxide, beta-glucose, guanidine acetic acid and beta-furose in serum of rats in group Y was lower than that in ED group, while the expression of VLDL and choline was higher (P<0.05).Conclusion The metabolic disorders of energy, amino acid and lipid appeared in the compound stress ED rat model, and the recovery of ED after Yimusake intervention may be promoted by regulating metabolism, inhibiting inflammation, oxidative stress and endothelial dysfunction.
