七叶胆苷ⅩⅦ对UV照射引起的小鼠内质网应激相关分子的影响

摘  要:目的:研究七叶胆苷ⅩⅦ对光老化小鼠皮肤组织中葡萄糖调节蛋白78(Glucose Regulated protein 78 kD,GRP78)、C/EBP的同源蛋白(C/EBP Homologous Protein,CHOP)、caspase-3内质网应激相关分子水平影响。探究七叶胆苷ⅩⅦ对光老化模型的疗效并探讨其作用机理。方法:将40只7周龄昆明雄性小鼠随机分为空白对照组、模型对照组、阳性药物对照组(VE组)、七叶胆苷组4组,将模型对照组、阳性药物对照组和七叶胆苷组小鼠进行光老化造模,空白对照组正常饲养。空白对照组和模型对照组给生理盐水,七叶胆苷组给相应剂量药液,阳性药物对照组给予维生素E滴剂,于造模前60 min灌胃给药。造模完成后,组织切片HE染色观察小鼠皮肤结构改变,采用ELISA法检测皮肤组织中GRP78、CHOP、caspase-3。结果:与空白对照组比较,模型对照组GRP78、CHOP、caspase-3表达水平明显上调,差异具有统计学意义(P<0.01);与模型对照组比较,七叶胆苷组GRP78、CHOP、caspase-3表达水平明显下调,差异具有统计学意义(P<0.01);与空白对照组相比,模型对照组表皮组织变薄,结构不完整,分层模糊不清;与模型组相比,阳性药物对照组(VE组)、七叶胆苷组表皮组织略有增厚,结构较为完整,各层细胞分界清楚。结论:七叶胆苷ⅩⅦ可能通过下调GRP78、CHOP、caspase-3等内质网应激相关分子的表达水平,从而一定程度上削弱内质网应激对皮肤组织的损伤,达到拮抗皮肤光老化的目的。Objective:To study the effect of GypenosideⅩⅦon the content of GRP78,CHOP and caspase-3 endoplasmic reticulum stress related molecules in skin tissues of photoaged mice.To explore the GypenosideⅩⅦeffect of light aging model and to explore its mechanism.Methods:Forty 7-week-old Kunming male mice were randomly divided into four groups:blank control group,model control group,positive drug control group(VE group),GypenosideⅩⅦgroup and model control group.The model of photoaging was made in the positive drug control group and the GypenosideⅩⅦgroup,while the blank control group was fed normally.The blank control group and the model control group were given normal Stroke-physiological saline solution,the GypenosideXVII group was given the corresponding liquid,and the positive drug control group was given vitamin E drops 60 min before establishment of the model.ELISA method was used to detect the content of GRP78,CHOP and caspase-3 in skin tissue,The changes of skin structure were observed by HE staining in tissue sections.Results:Compared with the blank control group,the expression of GRP78,CHOP and caspase-3 in the model control group was significantly up-regulated(P<0.01).Compared with the model control group,the expression of GRP78,CHOP and caspase-3 in the positive drug control group and the GypenosideⅩⅦgroup were significantly down-regulated(P<0.01).Compared with the blank control group,the model control group had thinner epidermal tissue,incomplete structure and fuzzy stratification.Compared with the model group,the epidermal tissue of the positive drug control group(VE group)and the GypenosideⅩⅦgroup was slightly thickened,with relatively complete structure and clear cell boundaries in each layer.Conclusion:GypenosideXVII may reduce the damage of endoplasmic reticulum stress to skin tissue by down-regulating the expression level,such as GRP78,CHOP and caspase-3,so as to antagonize the photoaging of skin.
