摘 要:以夏季采自河南省洛阳市、郑州市的实蝇幼虫、成虫为研究材料,采用DNA条形码技术,通过测定、比对线粒体DNA细胞色素氧化酶I(COI)基因片段序列,对其进行种类鉴定。结果表明,检测的实蝇样品种类为橘小实蝇(Bactrocera dorsalis)、具条实蝇(Zeugodacus scutellatus)和三点棍腹实蝇(Dacus trimacula),与相关种已知序列的相似度均在99.39%以上,其系统发育树明显与我国其他常见实蝇近缘种区分开。Tephritidae has a variety of species and causing serious damage to fruits, which is an important kind of agricultural and quarantine pests. Species identification of fruit flies is always relied on morphological identification of the adults, while its fragmentary adults, larvae and pupae cannot be accurately identified. The molecular identification of fruit fly based on DNA barcoding has attracted more attention of plant protection stations in recent years. Sequencing,alignment and blast with the mitochondrial cytochrome oxidase submit I(COI) gene were conducted on the fruit fly adults and larvae samples collected from Luoyang and Zhengzhou, Henan province in summer. The results showed that the specimen were Bactrocera dorsalis, Zeugodacus scutellatus and Dacus trimacula. The similarity was higher than 99.39%compared with the standard sequences, and these three species were clearly distinguished from other closely related species which often occurred in China shown by NJ phylogenetic tree. This study had practical significance for application of DNA barcoding techniques to achieve rapid and accurate identification of insect pests in plant protection stations.
