摘 要:采用硅胶柱层析、凝胶LH-20柱层析和半制备高效液相色谱等技术对樗蚕蚕沙中的化学成分进行分离纯化,得到9种单体化合物。利用质谱与核磁共振等波谱技术,分析并确定这9种化合物分别为吐叶醇、(3S,5R,6S,7E,9R)-5,6-环氧-3,9-二羟基-7-megastigmene、黄柏酮、neophellamuretin、黄柏苷、赪酮甾醇、赪酮甾醇3-O-β-D-葡萄糖苷和α-亚麻酸甲酯和叶绿醇。通过菌丝生长速率法测试化合物对4种植物病原真菌的抑菌活性,结果表明,吐叶醇对玉米大斑病菌(Exserohilum turcicum)、瓜果腐霉病菌(Pythium aphanidermatum)、水稻纹枯病菌(Thanatephorus cucumeris)和向日葵菌核病菌(Sclerotinia sclerotiorum)均有较为明显的抑制作用,其半最大效应浓度(EC50)分别为50.033μg/mL、54.016μg/mL、94.296μg/mL和47.144μg/mL;黄柏酮对水稻纹枯病菌和向日葵菌核病菌有较为明显的抑制作用,其EC50分别为126.811μg/mL和210.305μg/mL。Components in feces of Philosamia cynthia were separated and purified by silica gel column chromatography, Sephadex LH-20 chromatography and semi-preparative high-performance liquid chromatography, and 9 monomer compounds were obtained. Chemical constituents of 9 compounds were elucidated by spectroscopic technologies including mass spectroscopy(MS) and nuclear magnetic resonance(NMR), and were identified as vomifoliol,(3 S,5 R,6 S,7 E,9 R)-5,6-epoxy-3,9-dihydroxy-7-megastigmene, obacunone, neophellamuretin, phellamurin, clerosterol, clerosterol3-O-β-D-glucoside,α-linoleate and phytol. Their inhibitory activity against plant pathogenic fungi was determined by mycelium growth rate method. The results showed that vomifoliol could effectively inhibit the mycelium growth of Exserohilum turcicum, Pythium aphanidermatum, Thanatephorus cucumeris and Sclerotinia sclerotiorum, whose EC50 were 50.033 μg/mL, 54.016 μg/mL, 94.296 μg/mL and 47.144 μg/mL, respectively. Obacunone could effectively inhibit the mycelium growth of Thanatephorus cucumeris and Sclerotinia sclerotiorum, whose EC50 were 126.811 μg/mL and 210.305 μg/mL, respectively.
