当归四逆汤配合外敷药对冻伤模型大鼠的治疗作用

摘　　要：目的:分析当归四逆汤配合红花油、十滴水对冻伤模型大鼠的治疗作用及作用机制。方法:SD大鼠50只随机分为当归四逆汤治疗组、当归四逆汤配合红花油治疗组、当归四逆汤配合十滴水治疗组、冻伤模型组和正常对照组,每组10只。除正常对照组外,其余各组大鼠采用低温乙醇浸泡法制备大鼠左后足冻伤模型。当归四逆汤治疗组、当归四逆汤配合红花油治疗组、当归四逆汤配合十滴水治疗组大鼠灌胃当归四逆汤,正常对照组灌胃蒸馏水;当归四逆汤配合红花油治疗组、当归四逆汤配合十滴水治疗组大鼠左后足同时分别涂抹红花油、十滴水。比较各组大鼠足部外观,大鼠肿胀率,血清肿瘤坏死因子-β(tumor necrosis factor-β,TNF-β)、白细胞介素-6(interleukin-6,IL-6)水平。结果:冻伤模型大鼠出现不同程度的肿胀、溃烂及坏死,当归四逆汤、当归四逆汤配合红花油、十滴水可使症状缓解。与冻伤模型组比较,当归四逆汤治疗组、当归四逆汤配合红花油治疗组、当归四逆汤配合十滴水治疗组大鼠肿胀率显著降低(P<0.05),且当归四逆汤配合红花油组大鼠肿胀率较当归四逆汤组、当归四逆汤配合十滴水组显著降低(P<0.05)。与正常对照组比较,当归四逆汤治疗组、当归四逆汤配合十滴水治疗组和冻伤模型组大鼠TNF-β、IL-6水平显著升高(P<0.05);与冻伤模型组比较,当归四逆汤治疗组、当归四逆汤配合红花油治疗组、当归四逆汤配合十滴水治疗组大鼠TNF-β、IL-6水平显著降低(P<0.05),且当归四逆汤配合红花油组大鼠TNF-β、IL-6水平较当归四逆汤组、当归四逆汤配合十滴水组显著降低(P<0.05)。结论:当归四逆汤配合红花油较当归四逆汤、当归四逆汤配合十滴水对冻伤的治疗作用更为显著,其治疗冻伤的机制可能与调节TNF-β、IL-6的水平有关。Objective:To analyze the therapeutic effect and mechanism of Danggui Sini Decoction combined with Safflower oil and Ten drops(a popular medicine for summer ailments)on frostbite model rats.Methods:Fifty SD rats were randomly divided into the treatment group of Danggui Sini Decoction,Danggui Sini Decoction combined with Safflower oil treatment group,Danggui Sini Decoction combined with Ten drops treatment group,frostbite model group and normal control group,with 10 rats in each group.Except for the normal control group,the rats in the other groups were prepared by low temperature ethanol immersion method.The left feet of the rats in Danggui Sini Decoction treatment group,Danggui Sini Decoction combined with Safflower oil treatment group,Danggui Sini Decoction combined with Ten drops treatment group rats with Zhengzhi Sini Decoction,normal control group with distilled water;Angelica Sini Decoction with safflower,The oil treatment group,Danggui Sini Decoction and the Ten drips treatment group were simultaneously applied with Safflower oil and Ten drops.The appearance of the feet,the swelling rate of the rats,the levels of tumor necrosis factor-β(TNF-β)and interleukin-6(IL-6)were compared.Results:The rats in the frostbite model showed different degrees of swelling,ulceration and necrosis.The symptoms of Danggui Sini Decoction,Danggui Sini Decoction combined with Safflower oil and ten drops of water were alleviated.Compared with the frostbite model group,the swelling rate of Danggui Sini Decoction group,Danggui Sini Decoction combined with Safflower oil treatment group,Danggui Sini Decoction combined with Shishui treatment group was significantly lower(P<0.05),and Danggui Sini Decoction The swelling rate of the rats in the Safflower oil group was significantly lower than that in the Danggui Sini Decoction group and the Danggui Sini Decoction combined with the Shidou Shui group(P<0.05).Compared with the normal control group,the levels of TNF-βand IL-6 in Danggui Sini Decoction group,Danggui Sini Decoction combined w
