摘  要:饲料中过高比例植物油替代鱼油会引起肝脏炎症反应,养殖实验普遍存在周期长、价格昂贵以及干扰因素多等问题,离体实验部分弥补了养殖实验的缺点。植物油替代饲料中鱼油的本质是脂肪酸的替代。以往的研究发现豆油替代饲料中鱼油会引起炎症反应。为探究豆油对黄姑鱼肝脏细胞炎症影响,本实验分离并培养了黄姑鱼原代肝脏细胞。分别使用BSA(对照)、0.1mM二十碳五烯酸-牛血清白蛋白复合物(EPA-BSA)、0.1mM亚油酸-牛血清白蛋白复合物(LNA-BSA)孵育黄姑鱼原代肝脏细胞6h,用1.0mg·mL^-1LPS刺激细胞。结果表明:EPA与LNA相比,显著降低了促炎基因TLR2、TLR22、MyD88、TNF-α、IL-1β和IL-6的表达量(P<0.05)。本研究建立的黄姑鱼原代肝脏细胞模型可用于营养免疫研究,在黄姑鱼原代肝脏细胞中相同浓度的EPA相对LNA有更强抑炎效果,饲料中豆油替代鱼油引起的肝脏炎症反应可能是由于EPA含量减少,LNA含量升高引起的。Excessive proportion of vegetable oil in the feed instead of fish oil can cause liver inflammation. Culture experiments have may problems such as long cycle, high price and many interference factors. The in vitro experiment partially compensates for the shortcomings of the culture experiment. The essence of fish oil in vegetable oil substitute feed is the replacement of fatty acids. Previous studies have found that fish oil instead of soybean oil in feed lead to an inflammatory response. To explore the effects of soybean oil on liver cell inflammation in Nibea albiflora Richardson, N. albiflora liver cell was cultured in vitro. Liver cell was incubated with bovine albumin (BSA, the control), 0.1 mM eicosapentaenoic acid- bovine albumin complex (EPA-BSA) and 0.1 mM linoleic acid-bovine albumin complex (LNA-BSA) respectively for 6 hours, which was then stimulated with 1.0 mg·mL^-1 LPS. Results indicated that EPA significantly decreased gene expression level of TLR2, TLR22, MyD88, TNF-α, IL-1β and IL-6, when compared with LNA (P<0.05). Primary liver cell cultured in this study was suitable for nutritional immunology investigation;EPA exerted stronger anti-inflammatory effect than LNA in N. albiflora liver cell, liver inflammation caused by soybean oil instead of fish oil in feed may be caused by an decrease in EPA content and an increase in LNA content.
