摘 要:目的:观察7种商陆均一多糖对小鼠脾细胞增殖及细胞因子分泌的影响,探讨其对免疫调节的作用。方法:采用噻唑蓝(MTT)法检测7种商陆均一多糖对刀豆蛋白A(ConA)诱导的T淋巴细胞和脂多糖(LPS)诱导的B淋巴细胞增殖的影响;采用酶联免疫吸附法(ELISA)测定细胞上清液中IL-2、 IL-4、 IL-6、IFN-γ的含量。结果:商陆总多糖(PAP)在25 μg/mL、125 μg/mL的浓度下显著促进脾淋巴细胞增殖( P <0.05)。七种商陆均一多糖中,PAP-2、PAP-4、PAP-5、PAP-6及PAP-7对小鼠脾淋巴细胞增殖产生显著的促进作用;PAP在25 μg/mL、125 μg/mL的浓度下显著促进细胞因子IL-2、IL-4、IL-6和IFN-γ的分泌。七种商陆均一多糖中,PAP-2、PAP-4、PAP-5、PAP-6及PAP-7对小鼠脾细胞分泌细胞因子产生显著的增强作用。结论:商陆均一多糖PAP-2、PAP-4、PAP-5、 PAP- 6及PAP-7具有显著的免疫增强的作用。Objective: To observe the effects of 7 homogeneous polysaccharide from Phytolaccae on the proliferation of splenic lymphocytes and cytokines secretion of splenocytes in mice, and to explore their effects on immune regulation. Methods: MTT assay was used to detect the effects of 7 homogeneous polysaccharides from Phytolaccae on T lymphocyte proliferation induced by concanavalin A(ConA) and B lymphocyte proliferation induced by lipopolysaccharide(LPS). The contents of IL-2, IL-4, IL-6 and IFN-γ in cell supernatant were determined by enzyme-linked immunosorbent assay(ELISA). Results: The total polysaccharide of Phytolaccae (PAP) could significantly stimulate the proliferation of splenic lymphocytes at the concentration of 25 ug/mL and 125 ug/mL( P <0.05);among the 7 homogeneous polysaccharide, PAP-2, PAP-4, PAP-5, PAP-6 and PAP-7 significantly enhanced the proliferation of splenic lymphocytes. The total polysaccharide of Phytolaccae could significantly stimulate the secretion of IL-2, IL-4, IL-6 and IFN-γ in spleen cells at the concentrations of 25 ug/mL and 125 ug/mL;among the 7 homogeneous polysaccharide, PAP-2, PAP-4, PAP-5, PAP-6 and PAP-7 significantly enhanced the secretion of cytokines of spleen cells in mice. Conclusion: The homogeneous polysaccharide PAP-2, PAP-4, PAP-5, PAP-6 and PAP-7 from Phytolaccae have a significant immunological enhancement.
