摘  要:目的测定19个品种来源陈皮中多甲氧基黄酮川陈皮素和桔皮素的含量。方法色谱柱为Diamonsil C18柱(250 mm×4.6 mm,5μm),流动相为水(A)-甲醇(B)二元梯度洗脱(0~6 min时60%B,6~18 min时60%B→100%B),流速为1.0 mL/min,柱温为25℃,检测波长为326 nm。结果川陈皮素和桔皮素进样量分别在27.04~1 352.01 ng(r=0.999 9)和14.286~714.320 ng(r=0.999 6)范围内与峰面积线性关系良好,平均回收率分别为98.02%和98.60%,RSD为1.59%和2.75%(n=6);19批陈皮样品中川陈皮素和桔皮素含量测定结果分别为0.031%~0.968%,0.011%~0.546%,表明不同品种来源陈皮中多甲氧基黄酮含量有明显差异,江西南丰蜜桔、四川红橘和重庆江津大红袍含量较高,川陈皮素均在0.4%以上,桔皮素均在0.2%以上。结论经方法学验证,该方法可用于陈皮药材中川陈皮素和桔皮素的含量测定。Objective To determine the content of polymethoxylated flavones(nobiletin and tangeretin)in Pericarpium Citri Reticulatae from 19 cultivar origins.Methods The chromatographic column was Diamonsil C18 column(250 mm×4.6 mm,5μm),the mobile phase was water(A)-methanol(B)with binary gradient elution(0-6 min,60%B;6-18 min,60%B→100%B),the flow rate was1.0 mL/min,the column temperature was 25℃and the detection wavelength was 326 nm.Results The nobiletin and tangeretin showed good linear relationship with peak area in the ranges of 27.04-1 352.01 ng(r=0.999 9)and 14.286-714.320 ng(r=0.999 6),and the average recoveries were 98.02%%with RSD of 1.59%and 98.60%with RSD of 2.75%(n=6).The contents of nobiletin and tangeretin in the 19 batches of Pericarpium Citri Reticulatae were 0.031%-0.968%and 0.011%-0.546%.The results showed that the contents of polymethoxylated flavones had obvious differences among samples from different cultivar origins,which were relatively high from Citrus reticulata Blanco Kinokuni in Jiangxi,Citrus reticulata Zhuhong in Sichuan and Citrus reticulata Dahongpao in Jiangjin District of Chongqing(more than 0.4%of nobiletin and 0.2%of tangeretin).Conclusion According to the methodology validation,this method can be used for the content determination of nobiletin and tangeretin in Pericarpium Citri Reticulatae.
