中国新药创制的新内涵与新使命

摘　　要：疾病是因自稳调节紊乱而发生的异常生命活动,药物是辅助机体战胜疾病的调控物质,新药创制是阐明疾病发生发展机制、发现发展新的高效低毒药物、促进人类攻克各种疾病的活动。在历史的新起点上,新药创制的新内涵就是把握生命科学与其他学科在中国融合的新机遇,在疾病机制的解析、靶点的发现和确证、先导化合物的发现和优化、候选药物的确定、新药的临床研究及产业化等新药创制全价值链上进行整体布局,有效提升应对疾病的系统性、全局性和带动性能力,在推动构建人类命运共同体的征程中作出创新回答。目前人类疾病谱已发生重大变化,疾病防治从生物医学模式向"生理-心理-社会-环境"医学模式的逐渐转变,站在新的历史起点上,中国的新药创制迎来新机遇和新挑战。从多层次、多途径、多靶点的作用和调节层面对复杂疾病的治疗形成系统性的认知,进而从整体上协同各类优势技术资源,促成创新研究和关键技术的突破,提升中国新药创制体系的效率和效能,这是国家新药创制的新使命。The disease is an abnormal vital activity caused by the disorder of self-stabilization regulation,and the drug is a regulatory substance that assists the organism in overcoming the disease. The new drugs development is an activity that elucidates the mechanism of disease occurrence and development,discovers novel regulatory substance with high efficiency and low toxicity,and promotes humans to conquer various diseases. At the new historical starting point,the connotation of "new drugs development" is to seize the opportunity for the integration of bioscience and other disciplines,and to make plans for the value chain of innovation including the analysis of disease mechanisms,the discovery and confirmation of targets,the discovery and optimization of lead compounds,the determination of drug candidates,and the clinical research and industrialization of new drugs,so as to improve the systemic,overall and driving ability of disease response,and offer innovative solutions to promoting the building of a community of shared future for mankind. The human disease spectrum has changed greatly,and the disease prevention and treatment have gradually been transformed from a biomedical model to a "physio-psycho-socio-environment" medical model. The new drugs development in China is meeting new opportunities and challenges,and the new mission is to form a systematic understanding of the treatment of complex diseases from multi-channel,multi-target,multi-level functronal and regulatory perspective,coordinate various advantageous technological resources,promote breakthroughs in innovative researches and key technologies,and enhance the efficiency and potency of new drugs development system.
