摘 要:目的:探讨人脐带间充质干细胞(hUC-MSCs)移植对子宫内膜损伤大鼠的治疗作用及可能作用机制。方法:用无水乙醇诱导大鼠子宫内膜损伤模型,分为模型组、对照组、干细胞治疗组、雌激素治疗组。HE染色法观察子宫内膜组织学变化,测定子宫内膜厚度,免疫组化法检测血管内皮生长因子(VEGF)、雌激素受体(ERα)以及整合素β3表达。结果:对照组的子宫内膜厚度高于其他组(P<0.05),其余组间差异不明显。与对照组相比,模型组的VEGF、ERα、整合素β3表达均降低(P<0.05);与模型组相比,干细胞治疗组、雌激素治疗组的VEGF、ERα、整合素β3表达均升高(P<0.05);与雌激素治疗组相比,干细胞治疗组的VEGF、ERα、整合素β3表达均升高(P<0.05)。结论:hUC-MSCs移植和雌激素治疗均可不同程度地修复大鼠子宫内膜损伤组织,改善子宫内膜容受性,干细胞移植疗效优于单纯雌激素治疗。Objective:To investigate the therapeutic effect and possible mechanism of human umbilical cord mesenchymal stem cells(hUC-MSCs)transplantation on endometrial injury rats.Methods:The rat models of endometrial injury were established by absolute ethyl alcohol.The rats were randomly divided into four groups,the model group,the control group,the stem cell-treated group and the estrogen-treated group.The histological changes in the endometrium were observed by Hematoxylin-eosin staining.Endometrial thickness was measured.The expressions of vascular endothelial growth factor(VEGF),estrogen receptor(ER)and integrinβ3 in the endometrium were detected by Immunohistochemistry.Results:Endometrial thickness in the control group was higher than the other groups(P<0.05).There was no significant difference between the other groups.Compared with the control group,the expressions of VEGF,ERαand integrinβ3 in the model group were decreased(P<0.05).Compared with the model group,the expressions of VEGF,ERαand integrinβ3 were increased in the stem cell-treated group(P<0.05)and the estrogen-treated group(P<0.05).Compared with the estrogen-treated group,the expression levels of VEGF,ER,and integrinβ3 in the stem cell-treated group also increased(P<0.05).Conclusion:Human umbilical cord mesenchymal stem cell transplantation and estrogen treatment can repair the endometrial injury tissue of rats in different degrees.It may improve the endometrial receptivity.The effect of stem cell transplantation is superior to that of estrogen alone.
