摘  要:目的探讨甲基强的松龙(methylprednisolone,Meth)对大鼠脊髓损伤(spinal cord injury, SCI)后神经细胞凋亡及wnt/β-catenin信号通路的影响。方法将60只SD成年大鼠随机分为3组,即假手术(Sham)组、甲基强的松龙(Meth)组和生理盐水(Saline)组,每组各20只;假手术组仅需切开椎板不损伤脊髓,甲基强的松龙组和生理盐水组采用改良Allens法制作大鼠脊髓损伤模型后立即从尾静脉注射大剂量MP治疗,并在术后第1、2 d相同的时间点分别注射1次;采用尼氏染色(Nissl)观察组织形态变化;采用免疫荧光观察蛋白caspase-3表达水平;采用Western Blot检测各组wnt/β-catenin信号通路蛋白GSK-3β和β-catenin的表达水平。结果甲强的松龙组(Meth)第7 d SCI较生理盐水组明显减轻(P<0.01);Meth组抑制凋亡蛋白caspase-3表达水平明显下调(P<0.01);大鼠急性脊髓损伤(SCI)后第3、7 d,甲基强的松龙组呈现抑制蛋白GSK-3β表达(P<0.05),而蛋白β-catenin表达则明显上调(P<0.05)。结论大鼠急性脊髓损伤后MP可能通过激活wnt/β-catenin信号通路来促进大鼠运动功能恢复以及抑制急性脊髓损伤神经细胞的凋亡。Objective To investigate the effects of methylprednisolone(MP) on neuronal apoptosis and wnt/β-catenin signaling pathway after spinal cord injury(SCI) in rats.Methods Sixty adult SD rats were randomly divided into three groups: sham operation(Sham) group, methylprednisolone(Meth) group and saline group, with 20 rats in each group. The sham operation group only needed to incise the lamina without injuring the spinal cord. The rats in the methylprednisolone group and the saline group were injected with high dose MP through the caudal vein immediately after the model of spinal cord injury was made by modified Allens method. The rats in the methylprednisolone group and the saline group were injected once at the same time points on the first and second day after operation. Nissl staining was used to observe the morphological changes. Immunofluorescence was used to observe the expression level of caspase-3. Western Blot was used to detect the expression levels of wnt/β-catenin signaling pathway protein GSK-3β and β-catenin.Results Nissl staining showed that the injury of SCI tissue in methylprednisolone(Meth) group was significantly less than that in saline group(P<0.01). Immunofluorescence showed that Meth inhibited the down-regulation of apoptotic protein caspase-3 expression level(P<0.01). Western Blot showed that the expression of GSK-3β was inhibited in the methylprednisolone group on the 3 rd and 7 th day after SCI(P<0.05), while the expression of protein β-catenin was significantly up-regulated(P<0.05).Conclusion MP could promote motor function recovery and inhibit neuronal apoptosis after acute spinal cord injury by activating wnt/β catenin signaling pathway in rats.
