摘 要:目的分析不同浓度硝酸钕作用于发育期大鼠的行为学表现、病理学形态特征以及学习记忆的功能区海马组织的氧化损伤,评价硝酸钕对其学习记忆功能的影响及机制。方法 2018年3、9月选择SPF级SD性成熟大鼠32只,雌雄各半。随机将雌鼠分为低、中、高浓度硝酸钕组及阴性对照组,将雌雄大鼠按1∶1同笼进行交配,对受孕的大鼠持续染毒至仔鼠断乳,每组随机选取10只仔鼠,继续对仔鼠染毒21 d,观察各组仔鼠行为学表现、仔鼠海马的病理学形态以及海马的氧化损伤指标。结果 Morris水迷宫定位航行结果显示各组差异无统计学意义(P>0.05);而空间探索数据显示各组间比较差异有统计学意义(P<0.05),低浓度硝酸钕组穿越目标象限次数(20.40±5.68)、穿越原平台次数(6.50±3.02)以及时间百分比(42.25±10.07)高于阴性对照组(16.30±1.88、4.30±1.25、36.58±4.07)(P<0.05),高浓度硝酸钕组大鼠的时间百分比(33.56±4.16)低于阴性对照组(36.58±4.07)(P<0.05),路程百分比(25.59±4.84)低于阴性对照组(37.59±4.81)(P<0.01)。中高浓度组细胞间隙增宽,细胞排列稀疏紊乱,高浓度组部分细胞体积减小。海马的氧化损伤结果显示各组间海马SOD、MDA水平比较差异有统计学意义(P<0.01),两两比较显示中、高浓度硝酸钕组SOD水平低于阴性对照组(P<0.05),中、高浓度硝酸钕组SOD水平低于浓度硝酸钕组(P<0.01)。低浓度硝酸钕组MDA水平低于阴性对照组(P<0.05),中浓度硝酸钕组MDA水平高于阴性对照组及低浓度硝酸钕组(P<0.01)。结论长期接触中、高浓度硝酸钕可能抑制发育期大鼠学习记忆功能并对学习记忆功能区海马组织存在氧化损伤。Objective To analyze the behavioral and pathological features of neodymium nitrate at different concentrations in developing rats and the oxidative damage of the hippocampus in the functional area of learning and memory,and evaluated the effects and mechanism of neodymium nitrate on learning and memory. Methods A total of 32 SPF SD sexually mature rats were selected with half male and half female.Female rats were randomly divided into low,medium and high concentration of neodymium nitrate group and the negative control group,the male and female mating according to 1:1 with cage of rats,the pregnant rats were poisoned continuously until the offspring were weaned.10 young generation rats were randomly selected from each group and poisoned for 21 d,the behaviors,histopathological morphology and oxidative damage index of hippocampus were observed. Results The results of Morris water maze navigation showed that there was no significant difference among the groups(P>0.05).The data of space exploration showed that the differences among the groups were statistically significant(P<0.05).The times of crossing the target quadrant(20.40±5.68),the times of crossing the original platform(6.50±3.02) and the time percentage(42.25±10.07) in the low concentration neodymium nitrate group were higher than those in the negative control group(16.30±1.88,4.30±1.25,36.58±4.07)(P<0.05).The time percentage(33.56±4.16) and distance percentage(25.59±4.84) in the high concentration neodymium nitrate group were lower than those in the negative group(36.58±4.07,37.59±4.81)(P<0.01).Neodymium nitrate in medium and high concentration group showed cell atrophy,and a large number of nuclear pyknosis and inflammatory cell infiltration appeared in hippocampus.The oxidative damage of hippocampus results showed hippocampal SOD,MDA level compare differences among groups was statistically significant(P<0.01).Pairwise comparison showed SOD levels of medium and high concentration neodymium nitrate groups was lower than the control group(P<0.05) a
