ZD制剂对小鼠巨噬细胞cAMP、cGMP、PDE含量及cAMP/cGMP的影响

摘　　要：ZD制剂由川椒、茵陈、白芨等12味中草药经研磨、煎煮、浓缩再配以适当佐剂制成,是治疗马属动物肢蹄软组织损伤的经典处方,具有确实的临床疗效。cAMP和cGMP是真核细胞内的炎症反应调节因子,许多致炎因子通过破坏或改变炎症细胞内cAMP/cGMP的值起到致炎或抗炎作用。此次实验,目的:探讨ZD制剂对炎症过程中巨噬细胞中cAMP、cGMP、PDE含量及cAMP/cGMP比值的影响。方法:用脂多糖(LPS)刺激RAW264.7细胞,构建炎性细胞模型。用5种不同浓度的ZD制剂进行实验。采用ELISA法检测ZD制剂干预前后细胞cAMP、cGMP及PDE的变化,并计算cAMP/cGMP比值的变化。结果:ZD制剂可抑制模型细胞中PDE的表达并提高cAMP含量及cAMP/cGMP的比值;对cGMP未见明显影响。表明ZD制剂抗炎机制与抑制PDE表达,高cAMP含量及cAMP/cGMP比值密切相关。ZD is a classic prescription for curing the soft tissue injury in limbs of equus. cAMP and cGMP are inflammatory response regulators in eukaryocyte, and many inflammatory factors act as inflammatory or anti - inflammatory by destroying or changing the ratio of cAMP/cGMP in inflammatory cells. Objective: To investigate the effect of ZD on cAMP, cGMP, PDE and the ratio of cAMP/cGMP in macrophages during inflammation. Methods: RAW264. 7 cells were stimulated by lipopolysaccharide (LPS) and an inflammatory cell model was constructed. The experiment was carried out with 5 different concentrations of ZD preparation. ELISA method was used to detect the changes of cAMP,cGMP and PDE in ZD cells before and after intervention, and the change of cAMP/ cGMP ratio was calculated. Conclusion: ZD preparation can inhibit the content of PDE and increase the cAMP content and cAMP/ cGMP, and had no obvious influence on cGMP content, which showed that the anti-inflammatory mechanism of ZD preparation was closely related to the inhibition of PDE content and the increase of cAMP content and cAMP/cGMP ratio.
