地榆七柏汤灌肠治疗溃疡性结肠炎的实验研究

摘　　要：目的 观察地榆七柏汤灌肠对溃疡性结肠炎(UC)大鼠的治疗效果。方法 选80只清洁级Wistar雄性大鼠用于实验研究,空白组仅予盐水1mL/100g(鼠重)灌肠,其他组通过TNBS混合剂制备UC模型,之后模型组不给予药物治疗,柳氮磺胺吡啶(SASP)组和地榆七柏汤低(0.5g/mL)、中(1g/mL)、高(3g/mL)剂量组分别给予相应药物治疗。观察评估各组大鼠疾病活动指数(DAI)、血清IL-8和TNF-α水平以及结肠组织学变化。结果 造模后,SASP组和不同剂量地榆七柏汤组大鼠DAI评分随时间延长而不断降低(P<0.05),但SASP组和不同剂量地榆七柏汤组之间无统计学意义(P>0.05);造模后给予SASP或不同剂量地榆七柏汤灌肠均能明显降低TNF-α和IL-8水平(P<0.05),但SASP和不同剂量地榆七柏汤组之间无统计学意义(P>0.05);病理学检查显示,地榆七柏汤低剂量组能较好的修复结肠组织创面。结论 地榆七柏汤能改善溃疡结肠炎大鼠的疾病活动性、降低血清TNF-α、IL-8水平,促进结肠组织创面修复。Objective To observe the therapeutic effect of Diyuqibai decoction enema on ulcerative colitis (UC) rats. Methods Eighty clean Wistar male rats were selected for experimental study. The blank group was given saline enema of1 mL/100 g (rat weight) only, the other groups were prepared UC model through TNBS mixture. After that, the model group was not given drug treatment. Sulfasalazine (SASP) group and Diyuqibai decoction low(0.5 g/mL), medium(1 g/mL) and high(3 g/mL) dose groups were given corresponding drug treatment. The disease activity index (DAI), serum levels of IL-8 and TNF-α, HE staining of colon tissue were observed and evaluated. Results After modeling, DAI scores of rats in SASP group and different doses of Diyuqibai decoction group decreased when time increase ( P <0.05), but there was no significant difference between them ( P > 0.05). After modeling, SASP or different doses of Diyuqibai decoction could significantly reduce the levels of TNF-αand IL-8 ( P <0.05), but there was no significant difference between them ( P >0.05). Pathological examination showed that the low dose Diyuqibai decoction group could repair the wound of the colon wound better. Conclusion Diyuqibai decoction can improve the DAI score of ulcerative colitis rats, reduce serum levels of TNF-αand IL-8, and promote the repair of colonic wounds.
