摘  要:为探明旅游开发以及旅游干扰活动对三亚临春岭森林公园土壤细菌群落丰度和多样性的影响,为热带地区森林资源的保护与合理开发提供依据,研究选取三亚临春岭森林公园为调查对象,沿登山步道垂直方向由近及远选取三个区域,游人干扰区(0~3m)、景观绿化区(10~30m)、人工成熟林区(100~150m),分别提取土壤总DNA并利用Illumina高通量测序技术分析土壤细菌的群落构成和多样性。三个区域表层土壤共获得134713条序列,OTU总数为1936个,其中三个样地的共有OUT数目为424个,约占OUT总数的21.9%。三个区域的土壤细菌可分为13个门类,变形菌门(Proteobacteria)在三个区域中均占绝对优势。人工成熟林区土壤微生物多样性最高,而游人干扰区和景观绿化区微生物分布更相近。Taking Sanya Linchunling Forest Park as example,we studied the effect of tourism development and tourism trampling disturbance on diversity and richness of soil bacteria ,in order to offer a foundation for the development of forest tourism in tropic regions. In vertical directions along the mountain trails,the area can be divided into three zones,tourist disturbance area (0~3 m),Landscape green area (10~30 m) and mature artificial forest region (100~ 150 m ). The total soil DNA was extracted and sequenced using Illumina s sequencing technology for soil microbial diversity and richness.In the three zones,totally 134 713 sequences were obtained and divided into 1 936 OTUs,homologous OTU number of the three zones is 424 which is about 21.9 % of total OTUs.The soil bacteria in three zones which belonged to 13 phylums,and Proteobacteria make up absolute predominance in the three zones.Soil bacteria diversity in pristine forest region was far greater than the other two zones and the bacterial species composition of Landscape green area and artificial forest region were more similar.
