摘 要:目的建立测定大鼠血浆中左氧氟沙星质量浓度的超高效液相色谱(UPLC)法方法色谱柱为Waters ACQUITY UPLC BEH C18柱(50mm×2.1mm,1.7μm),流动相为0.05mol/L磷酸二氢钾缓冲液-乙腈(75∶25,V/V),流速为0.3mL/min,柱温为30℃,进样量为1μL,检测波长为294nm。结果左氧氟沙星质量浓度在0.20~15.00μg/mL范围内与对照品和内标的峰面积之比线性关系良好,r=0.9994(n=7)。检测限为0.20μg/mL,日内、日间精密度及稳定性试验的RSD均小于8%(n=6)。结论该方法专属性强,灵敏度高,准确性好,检测速度快,适用于大鼠血浆中左氧氟沙星的药代动力学研究。Objective To establish a UPLC method for concentration determination of levofloxacin in rat plasma.Methods The chromatographic column was Waters ACQUITY UPLC BEH C18 column(50 mm×2.1 mm,1.7μm),the mobile phase was 0.05 mol/L potassium dihydrogen phosphate buffer-acetonitrile(75∶25,V/V),the flow rate was 0.3 mL/min,the column temperature was 30℃,the sample size was 1μL and the detection wavelength was 294 nm.Results The levofloxacin showed good linear relationship with peak area in the range of 0.20-15.00μg/mL,r=0.999 4(n=7).The limit of detection(LOD)was 0.20μg/mL and RSDs of the intra-precisions and inter-precisions and stability tests were all lower than 8%(n=6).Conclusion The method is specific,sensitive,accurate and fast,which is suitable for studying the pharmacokinetics of levofloxacin in rat plasma.
