摘 要:目的检测干扰素-γ(IFN-γ)预处理骨髓间充质干细胞(BMSCs)中PD-L1的表达及对初始T细胞分化的作用,同时探讨同种异体免疫调节的作用机制。方法采用不同浓度的IFN-γ预处理BMSCs,检测PD-L1的表达量,并分析IFN-γ预处理BMSCs对反应激活的T细胞增殖和分化的影响。结果用IFN-γ预处理BMSCs可以抑制混合淋巴细胞反应激活的T细胞的增殖,其在加入PD-L1阻断剂后消失。结论BMSCs的免疫调节作用受IFN-γ浓度的影响,其不仅抑制T细胞的活化,还诱导同种异体抗原激活的T淋巴细胞分化为Treg。Objective To detect the expression of PD-L1 in bone marrow mesenchymal stem cells (BMSCs) pretreated with interferon-γ(IFN-γ) and its effect on the differentiation of initial T cells, and to explore the mechanism of alloimmune regulation. Methods BMSCs were pretreated with different concentrations of IFN-γ to detect the expression of PD-L1, and the effects of IFN-γ pretreatment on the proliferation and differentiation of reactive T cells were analyzed. Results Pretreatment of BMSCs with IFN-γ inhibited the proliferation of T cells activated by mixed lymphocyte reaction and disappeared after adding PD-L1 blocker. Conclusion The immune regulation of BMSCs is influenced by IFN-γ concentration, which not only inhibits the activation of T cells, but also induces T lymphocytes activated by alloantigen to differentiate into Treg.
