摘 要:从黄河三角洲东营胜利油田附近石油污染的土壤中分离、筛选到菲降解菌共6株,并对其中降解率相对较高的菌株PAHs-1进行了生物学特性的研究,包括菌体形态特征、培养特征及生理生化特征的初步研究.通过形态观察、生理生化指标及16SrDNA 序列分析,鉴定该菌株为短芽孢杆菌属.研究所筛选的PAHs-1能够以多环芳烃菲为唯一碳源和能源,7d内对菲(培养液浓度为50mg/L)的降解率达到62.09%.并通过试验确定了PAHs-1对菲的最佳降解pH 为7.另外,研究中一系列试验表明随着外源碳源葡萄糖的加入,PAHs-1对菲的降解率呈微小的增大趋势,而随着菲浓度的增加,PAHs-1对菲的降解率呈减小趋势.In this study,six strains of phenanthrene degrading bacteria were isolated and screened from oil-polluted soid near dongying Shengli oilfield in the Yellow River delta.The biological characteristics for relatively high degradation rate of PAHs-1 strain were studied,including preliminary morphological studies,the physiological and biochemical characteristics of culture and bacteria.The strain was identified as short bacillus by morphological observation,physiological and biochemical indexes and 16S rDNA sequence analysis.PAHs-1 screened in this study can be used as the only carbon source and energy source to degrade phenanthrene(medium concentration of 50 mg/L)at a rate of 62.09%within 7 days.The optimum pH for PAHs-1 degradation of phenanthrene was 7.In addition,a series of experiments in this study showed that,with the addition of exogenous carbon source glucose,the degradation rate of phenanthrene showed a slight upward trend,while with the increase of phenanthrene concentration,the degradation rate of phenanthrene showed a downward trend.
