摘  要:从黄野螟(Heortia vitessoides Moore)转录组文库中成功筛选到具有完整ORF框的黄野螟中性肽链内切酶基因(NEP)序列,命名为HvNEP(登录号:MH298329)。该序列ORF框长2 310 bp,共编码769个氨基酸。同源比对分析结果表明,HvNEP蛋白氨基酸序列与棉铃虫(Helicoverpa armigera Hubner)NEP蛋白氨基酸序列的同源性最高,达79%。RT-qPCR结果显示,HvNEP在黄野螟成虫中表达量最高,为卵(对照)的113.77倍。HvNEP在黄野螟不同组织间存在表达差异,幼虫阶段在头部表达量最高,为体壁(对照)的5.28倍。成虫阶段,HvNEP在翅中表达量最高,为腹(对照)的11.06倍,在足中最低(0.11倍)。分析黄野螟NEP时空表达动态发现,HvNEP可能与昆虫行为的调控及神经系统的完善有关。在60 ng/μl和120 ng/μl的20-羟基蜕皮激素(20E)胁迫下,HvNEP表达量均高于对照,表明HvNEP作为蜕皮激素的次级应答基因,其表达受20-羟基蜕皮激素调节。The full-length cDNA sequence of H.vitessoides neprilysin gene(NEP)was screened from the transcriptome library,and named HvNEP(GenBank:MH298329).The sequence open reading frame(ORF)was 2 310 bp in length and encoded 769 amino acids.Homology alignment analysis results showed that the amino acid sequence of HvNEP protein had the highest homology with that of the Helicoverpa armigera Hubner NEP protein(79%).According to the results of RT-qPCR analysis,the expression of HvNEP in adult was highest,which was 113.77 times of that in eggs(control).The expression of HvNEP was different among different tissues of H.vitessoides.The highest expression in the larvae was 5.28 times of that in the body wall(control).In the adult stage,the expression of HvNEP was the highest in the wings(11.06 times of that in the abdomen)and the lowest in the feet(0.11 times of that in the abdomen).Temporal and spatial dynamics expression of HvNEP indicated that it might be involved in the regulation of insect behavior and the improvement of nervous system.Under the stress of 60 ng/μl and 120 ng/μl 20-hydroxyecdysone(20E),the expression of HvNEP was higher than that of the control,indicating that HvNEP,as a secondary response gene of ecdysing hormone,was regulated by 20E.
