摘  要:目的:探讨青砖茶水提物对高脂饮食诱导的C57肥胖小鼠的抑制作用及其潜在机制。方法:50只5周龄的健康雄性C57小鼠随机分为5组:对照组、模型组、青砖茶水提物低、中、高剂量组,每组10只。对照组给予普通饲料喂养,模型组给予高脂饲料喂养,青砖茶水提物低、中、高剂量组在高脂饲料喂养的基础上分别给予低剂量[JP2](75mg/kg)、中剂量(150mg/kg)和高剂量(300mg/kg)[JP]青砖茶水提物。20w后测量小鼠体重、体长,计算肥胖指数(LEE指数),检测血清中总胆固醇和甘油三酯水平,骨骼肌中丙二醛(MDA)、活性氧(ROS)和蛋白质羰基化(PC)的含量,超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GPx)、过氧化氢酶(CAT)和谷胱甘肽还原酶(GR)的活性。采用RT-PCR和Westernblot检测核因子E2相关因子2(Nrf2)信号通路活性及其下游抗氧化因子的表达水平。结果:与对照组比较,模型组小鼠体重、LEE指数、血脂及骨骼肌中MDA、ROS和PC显著升高,抗氧化酶SOD、GPx、CAT和GR的活性下降(均P<0.05)。与模型组比较,青砖茶水提物干预20w后,小鼠体重、LEE指数、血脂及骨骼肌中MDA、ROS和PC降低,抗氧化酶SOD、GPx、CAT和GR活性水平升高,并呈剂量依赖性。高剂量青砖茶水提物干预后,Nrf2/血红素加氧酶-1(HO-1)信号通路及下游抗氧化因子谷胱甘肽S-转移酶(GST)、γ-谷氨酰半胱氨酸连接酶(γ-GCL)和磷酸酰胺腺嘌呤二核苷酸醌氧化还原酶-1(NQO-1)的蛋白和mRNA表达较模型组均显著增加(均P<0.05)。结论:青砖茶水提物可明显改善肥胖和脂代谢紊乱,其机制可能与Nrf2/HO-1信号通路活化有关。Objective: To explore the effect and mechanism of qing brick tea water extract on obesity and oxidative stress induced by high-fat diet.Methods: Fifty male C57 mice were randomly divided into five groups, including the control group, the model group, the low-, moderate- and high- dose of qing brick tea water extract group, with 10 mice in each group.Mice in the control and the model group were given normal and high fat diet, respectively.In addition to high fat diet, mice in the low-, moderate- and high- dose of qing brick tea water extract group were administrated with qing brick tea water extract at a dose of 75 mg/kg, 150 mg/kg and 300 mg/kg, respectively.At 20 weeks after intervention, the body weight, body length, and LEE index were measured.The levels of total cholesterol and triglyceride in serum were measured using commercial kits.The content of malondialdehyde (MDA), reactive oxygen species (ROS) and protein carbonyl (PC), as well as the activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT), glutathione reductase (GR) in skeletal muscle were detected.Besides, the expression of nuclear factor E2-related factor 2 (Nrf2) and its downstream antioxidant factors were also made further evaluation.Results: Compared with the control group, mice in the model group performed obvious rising tends in body weight, LEE index, serum lipid, MDA, ROS and PC in skeletal muscle, and significant decline of antioxidant enzymes activities in the skeletal muscle, including SOD, GPx, CAT and GR (All P <0.05).However, qing brick tea water extract displayed dose-dependent inhibitory effects on body weight, LEE index, serum lipid, MDA, ROS and PC in skeletal muscle, while enhanced activities of anti-oxidative enzymes, including SOD, GPx, CAT and GR, when compared with the model group.Moreover, the activity of Nrf2/ heme oxygenase-1(HO-1) pathway and its downstream antioxidant factors were all increased by qing brick tea water extract, containing glutathione S-transferase (GST),γ-glutamyl cysteine l
