摘　　要：目的探讨Toll样受体在不同转移能力人乳腺癌细胞株中的差异表达及意义。方法分别选取高转移能力人乳腺癌细胞株MDA-MB231与低转移能力人乳腺癌细胞株MCF-7,采用RT-PCR、实时荧光定量PCR及细胞免疫荧光检测测定其蛋白水平及TLR1-TLR10mRNA水平,比较两株细胞TLRs表达情况及差异性。结果人乳腺癌细胞株MCF-7mRNA、MDA-MB-231mRNA、蛋白水平均存在TLRs广泛表达;两种细胞株TLR4、TLR6、TLR8及TLR9mRNA水平表达存在明显的差异性(P<0.05);MDA-MB-231的TLR4、TLR6、TLR7及TLR5水平明显高于MCF-7(P<0.05)。结论TLRs在不同转移能力人乳腺癌细胞株中表达广泛但水平存在差异,其中TLR4受体差异表现最为明显,考虑TLRs表达差异与乳腺癌细胞转移能力相关。Objective To investigate the differential expression and significance of Toll-like receptors in human breast cancer cell lines with different metastatic ability. Methods High-metastatic human breast cancer cell line MDA-MB231 and low metastatic human breast cancer cell line MCF-7 were selected, and their protein levels and TLR1-TLR10 mRNA level were determined by RT-PCR, real-time fluorescent quantitative PCR and cellular immunofluorescence assay.The expression and difference of TLRs in the two cells was compared. Results The expression of TLRs was widely expressed in human breast cancer cell line MCF-7 mRNA and MDA-MB-231 mRNA and protein expressions. There are significant differences in the expression of TLR4, TLR6, TLR8 and TLR9 mRNA levels in the two cell lines(P<0.05). The levels of TLR4, TLR6, TLR7 and TLR5 in MDA-MB-231 are significantly higher than those in MCF-7 (P<0.05). Conclusion TLRs are widely expressed in different metastatic human breast cancer cell lines, while the expression levels are different. The difference of TLR4 receptors is the most obvious, considering that the difference of TLRs expression is related to breast cancer cell metastasis ability.
