摘 要:用荧光和紫外光谱法研究了苏丹Ⅳ与DNA在含金属离子环境中的相互作用.结果表明:DNA及金属离子均导致苏丹Ⅳ的内源荧光猝灭,猝灭机制均为静态猝灭.运用Scatchard方程对实验数据进行处理分析,求得在有无金属离子存在下苏丹Ⅳ与DNA相互作用的结合常数和结合位点数.讨论了金属离子对苏丹Ⅳ与DNA相互作用的影响.The interation between Sudan Ⅳ and herring sperm DNA in the presence of metal ions was investigated by fluorescence and ultraviolet spectroscopy. The results showed that both DNA and metal ions cause the quench of the fluorescence intensity of Sudan Ⅳ in a static quenching mechanism. The binding constants and sites in the absence and presence of metal ions were obtained by scatchard equation . The effects of metal ions on the interaction of Sudan Ⅳ with DNA were also discussed.
