摘 要:目的明确脐带间充质干细胞(UC-MSCs)对2型糖尿病(T2DM)db^-/db^-小鼠的临床疗效及安全有效的移植途径,旨在为临床应用提供实验与理论数据。方法db/m和db^-/db^-小鼠各10只,每种小鼠各分为2组,db/m对照组,db/m+UC-MSCs组,db^-/db^-模型组,db^-/db^-+UC-MSCs组,5只/组。对照组和模型组给予1mL生理盐水腹腔注射;db/m+UC-MSCs和db^-/db^-+UC-MSCs组腹腔注射UC-MSCs为1×10^7个细胞/只鼠,总液体量为1mL。每周固定时间记录餐后血糖与体重,计算各组小鼠生存率。结果与db-/db-模型组小鼠相比,腹腔途径移植UC-MSCs后,db^-/db^-小鼠的血糖水平明显下降,与非移植db^-/db^-小鼠比较差异有显著性,对db/m小鼠无影响;移植与非移植小鼠体重无明显变化。db^-/db^-+UC-MSCs组小鼠出现部分死亡,生存率为75%,其它组小鼠生存率均为100%。结论UC-MSCs腹腔移植途径可有效控制T2DM小鼠血糖变化,对体重变化无明显影响。可为临床治疗提供参考。Objective To investigate the clinical efficacy of UC-MSCs on T2DM in db ^-/db ^- mice, and look for a kind of safe and effective transplantation pathway, so as to provide experimental and theoretical data for clinical application. Methods 10 db/m and10 db ^-/db ^- mice were randomly divided into 4 groups: db/m normal control group, db/m+UC-MSCs group, db ^-/db ^- model group, db ^-/db ^-+UC-MSCs group, 5 mice/group. 1 mL saline were infected by intraperitioneal in control group and model group, mice from db/m+UC-MSCs group and db ^-/db ^-+UC-MSCs group were transplanted UC-MSCs 1×10^ 7/mice. Postprandial blood glucose and body weight were recorded at a fixed time every week, and survival rate in each group mice were calculated. Results The blood glucose in db ^-/db^- mice was controlled effectively by abdominal transplantation, but had no significantly effect on body weight. To be mentioned, db ^-/db^- mice transplanted UC-MSCs by intraperitoneal injection appeared death, thus clinical application should be cautious. Conclusion Intraperitoneal injection UC-MSCs can effectively control blood glucose in db ^-/db ^- mice, but had no obviously effect on body weight. Although db ^-/db^- mice appeared death transplanted by intraperitoneal injection, the mortality was low, which could be used as a clinically effective UC-MSCs transplantation pathway after comprehensive factor evaluation.
