摘  要:以北京市奥体公园再生水补水人工湿地净化系统芦苇根际样品为研究对象,采用末端限制性片段长度多态性技术(TerminalRestriction Fragment Length Polymorphism, T-RFLP),通过传统多样性指数分析与基于多酶切综合比对细菌群落多样性变异分析和单酶切T-RFs 类群丰度显著变化特征分析相结合的方法对芦苇根际细菌群落多样性空间差异进行分析,并借助单因素方差分析(one-wayANOVA)和Spearman 相关分析等多元统计方法对再生水人工湿地净化系统各功能区细菌群落种属差异进行判别,分析人工湿地不同空间尺度的水质净化特征。结果表明,基于全模型判别分析法识别奥林匹克公园典型再生水补给湿地不同水质净化功能区具有显著差异的水质指标为pH、DO、ORP、TN 和NH4^+-N 5 项。传统多样性指数分析方法未能对功能区间细菌群落多样性空间差异做出有效分析,而基于多酶切综合比对细菌群落多样性变异分析结果表明复合垂直流湿地和植物氧化塘的细菌群落多样性明显低于混合生态功能区,pH、DO、Cl^-、TN 和NO3^--N 是导致人工湿地净化系统芦苇根际细菌群落多样性空间变异主要因素。β-proteobacteria、Cyanobacteria和Actinobacteria 丰度在各类群落间变化显著。再生水补水中有机污染物的降解主要发生在复合垂直流人工湿地和植物氧化塘。Reeds rhizosphere samples from the reclaimed water recharged constructed wetland purification system in Beijing Olympic Sports Park were taken as research objects. Terminal restriction fragment length polymorphism (T-RFLP) was used to analyse the conventional diversity index of bacterial community so as to find its spatial variation in reeds rhizosphere. One-way ANOVA and Spearman correlation analyses were also used to discriminate the significant changes in the abundance of different groups of bacterial community. Based on the full model discriminant analysis method, following water quality indicators have been identified with significant differences in different water purification functional areas, i.e., the indicators of pH value, DO, ORP, total nitrogen, and ammonium nitrogen. It revealed that those five indicators could be used to characterise the differences in water quality among different functional areas. Significant changes in the abundance of β-proteobacteria, Cyanobacteria and Actinobacteria in various communities were found. It indicated that pH value, DO, chlorides, total nitrogen and nitrate-nitrogen contents might be the direct factors to cause the spatial variation in the diversity of bacterial community. The degradation of organic pollutants in recharged reclaimed water would mainly occur in the composite vertical-flow constructed wetland.
