摘  要:采用PCR技术对乌克兰鳞鲤、德国镜鲤、框鳞镜鲤、红镜鲤和州河鲤共97尾个体的线粒体DNA D-loop及邻近区段进行了扩增,经测序后获得碱基顺序排列清晰的DNA片段,长度分别为1342、1343、1344 bp。共得到8种单倍型(GenBank序列号为MG786481~MG786483,MG786485~MG786487,KC292935,KC292936)。使用Mega 6.0软件计算出5个群体的组内及组间遗传距离,并构建邻接系统树。线粒体DNA D-loop及邻近区段、单独D-loop区段的碱基序列分析结果表明,单倍型Ⅰ至Ⅳ为欧洲血统,碱基差异位点数较多,变异程度较高;单倍型Ⅴ至Ⅷ为亚洲血统,碱基差异位点数较少,变异程度较低。在线粒体DNA D-loop及邻近区段内,德国镜鲤组内平均遗传距离为0.00462,核苷酸多样性指数为0.00458,平均核苷酸差异性为6.150,遗传多样性最大;州河鲤组内的平均遗传距离为0.00065,核苷酸多样性指数为0.00065,平均核苷酸差异性为0.870,遗传多样性最低。In this study,the mitochondrial DNA (mtDNA) D-loop and its adjacent region were amplified in 97 samples including of Zhouhe carp,mirror carp,Germany mirror carp,red mirror carp and Ukraine scale carp by PCR. The DNA fragment lengths were found to be 1342 bp,1343 bp and 1344 bp. The sequences of 8 haplotypes had submitted to GenBank,and the sequence numbers were MG786481-MG786483,MG786485-MG786487,KC292935 and KC292936. The genetic distance and neighbor-joining (NJ) phylogenetic tree analysis were performed by Mega 6.0 software. Through the analysis of base sequence in mtDNA D-loop and its adjacent region or D-loop region,the results showed that the haplotypesⅠ to Ⅳ were European descent,and that there were many points of base difference,with low variation extent. The haplotypes Ⅴ to Ⅷ were Asian descent,and that there was little point of base difference,with high variation extent. In mtDNA D-loop and its adjacent region,the average genetic distance within the Germany mirror carp group was 0.00462,with nucleotide diversity of 0.00458,average nucleotide differences of 6.150,and the maximal genetic diversity,the average genetic distance within the Zhouhe carp group of 0.00065 ,nucleotide diversity of 0.00065,with average nucleotide differences number of 0.870,the minimal genetic diversity among the five types of common carp.
