摘 要:目的检测宫颈脱落细胞中人类染色体端粒酶基因(telomerase human gene,hTERC)和人髓细胞增生原癌基因(myelocytomatosis oncogene,C-MYC)的表达,探讨在宫颈病变的筛查和诊断中其所具有的实用价值。方法宫颈癌筛查患者均行宫颈脱落细胞中hTERC基因和C-MYC基因阳性表达的检测。结果(1)在正常组,低度宫颈上皮内瘤变组,高度宫颈上皮内瘤变组,宫颈浸润癌组,hTERC基因异常表达,且各组间差异比较有统计学意义(P<0.05);(2)在正常组,低度宫颈上皮内瘤变组,高度宫颈上皮内瘤变组,宫颈浸润癌组,各组间C-MYC基因的表达率比较存在差异,且有统计学意义(P<0.05)。结论(1)hTERC基因在正常组及不同级别的宫颈病变组均异常表达,且阳性率随着病变程度的增加而上升,可作为检测子宫颈癌癌前病变的生物遗传学指标,可期望成为筛查早期宫颈癌的方法之一;(2)C-MYC基因在正常组及不同级别的宫颈病变组异常表达,且阳性率随着病变程度的增加而增加,可作为检测子宫颈癌癌前病变的指标,且可期望成为早期宫颈癌筛查中一种新的检测手段。Objective To detect the express of telomerase human gene(hTERC) and myelocytomatosis oncogene(C-MYC) in cytological specimens of cervix and to explore their value in screening and diagnosing of cervical Lesions.Methods According to histological biopsy form October 2010 to December 2011,137 patients were divided into four groups-normal(n=32),CINⅠ(n=66),CINⅡ/CINⅢ(n=31),ICC(n=8).Fluorescence in situ hybridization(FISH) was used to detect the expression of hTERC and C-MYC.Results (1)CINⅠ,CINⅡ/Ⅲ,and ICC groups,there were significant differences in hTERC expression between every two groups (P<0.05).(2)CINⅠ,CINⅡ/Ⅲ,and ICC groups,there were significant differences in C-MYC expression between every two groups(P<0.05). Conclusions(1)Abnormal expression of hTERC exists in normal,CINⅠ,CINⅡ/Ⅲ,and ICC patients,which significantly increases during malignant development.It may be biogenetics monitor indexes of cervical precancerosis and will be screening markers for cervical cancer.(2)Abnormal expression of C-MYC exists in CINⅠ,CINⅡ/Ⅲ,and ICC patients,which significantly increases during malignant development.It may be biogenetics monitor indexes of cervical precancerosis and will be screening markers for cervical cancer.
