摘  要:为提高兰州百合再生植株效率,保存优质种质资源,以兰州百合鳞片为外植体,MS为基本培养基,就初代有无不定芽产生对继一代试管苗生长的影响以及α-萘2酸(NAA)和6-苄氨基腺嘌呤(6-BA)浓度在0.1~1.0mg/L范围内对继一代、继二代和继三代试管苗生长发育的影响进行了研究。结果表明:(1)在继一代中,3种激素浓度处理表现为有不定芽处理外植体获得率、转绿率均远高于无不定芽处理;不管培养多长时间,无不定芽处理都无生命体征,死亡率亦远高于有不定芽处理。因此初代诱导产生不定芽的小鳞片是继一代培养的最佳材料。(2)继一代以MS+0.5mg/LNAA+0.5mg/L6-BA为最佳培养基,诱导培养33d后,生叶率、小鳞茎数和叶片数分别为69.52%、6.67个和14.50片,但死亡率仅为13.02%;继二代以MS+0.1mg/LNAA+1.0mg/L6-BA为最佳培养基,生叶率、叶片数和小鳞茎数在最高时分别达到91.18%、21.00片和7.67个;继三代培养以MS+0.1mg/LNAA+0.75mg/L6-BA为适宜培养基,生叶率和发芽率在各阶段均为100%和0%。In order to improve the regenerated efficiency of Lanzhou lily and preserve the high- quality germplasm resources,the author took the scale of Lanzhou lily as explant,used MS as the basic culture medium.An experiment was conducted to investigate the effects with or without adventitious bud on the growth of the first subculture as well as the roles of the concentrations(0.1~1.0 mg/L) of NAA and 6-BA on the growth of the first-,second- and third- subculture.The results showed that:(1) The rate of explant acquisition and the rate of turn green were much higher than the first-subculture without adventitious buds under the three hormone concentrations.There was no vital sign in the treatment of without adventitious bud no matter the length of time culture.Moreover,the mortality rate was much higher than that with adventitious bud.Therefore,it is the best material that the initial generation can induce adventitious buds in the first subculture.(2)The best medium for the first subculture was MS+NAA 0.5 mg/L+6-BA 0.5 mg/L.After 33 days of induction culture,the leaf producing rate,number of leaves and small bulbs reached 69.52%,6.67 and 14.50,respectively,but the mortality rate was only 13.02%.MS+NAA 0.1 mg/L+6-BA 1.0 mg/L was the best medium for the second subculture,the leaf producing,number of leaves and small bulbs could reach 91.18%,21 and 7.67,respectively,at the highest point.In the third subculture MS+NAA 0.1 mg/L+6-BA 0.75 mg/L was used as the suitable medium.the leaf producing and germination rate were 100.00% and 0.00% at each stage,which accorded with the growth law of test-tube seedlings at later stage.
