炮姜对虚寒性出血大鼠模型的影响

摘　　要：目的:观察炮姜水煎液对虚寒性出血大鼠模型的治疗作用。方法:分别建立脾胃虚寒出血大鼠模型及虚寒性子宫出血大鼠模型。利用米醋、乙醇联合吲哚美辛建立脾胃虚寒性出血大鼠模型。将大鼠随机分为空白组、阳性组、炮姜高、中、低剂量组,连续给药7 d。取胃、大肠、小肠做病理切片,酶联免疫吸附法测定胃动素(motilin,MTL)、胃泌素(gastrin,GAS)的含量,测定凝血四项。利用米非司酮配伍米索前列醇联合冰食水建造虚寒性子宫出血大鼠模型。将大鼠随机分为空白组、阳性组、炮姜高、中、低剂量组,连续给药7 d。末次给药后处死,取子宫做病理切片,测定凝血四项,并检测每日出血量。结果:炮姜各剂量组均能够显著改善脾胃虚寒出血模型、虚寒性子宫出血模型大鼠的外观状态,提高进食量、饮水量,显著改善凝血四项指标,缩短凝血时间;脾胃虚寒出血模型大鼠MTL、GAS的含量显著升高,病理切片显示与模型组相比组织损伤较轻,病理改变轻且浅;虚寒性子宫出血模型大鼠出血量减少。结论:炮姜有温经止血的作用。Objective : To observe the therapeutic effect of decoction of processed ginger on rat model of deficient cold of bleeding. Methods: Rat models of spleen-stomach deficiency cold bleeding and uterine bleeding due to deficiency cold were established. The rat model of spleen?stomach deficiency cold bleeding was established by rice vinegar and ethanol combined with indomethacin. Rats were randomly divided into blank group,positive group, high,medium and low dose groups of decoction of processed ginger, and were administered continuously for 7 days. Pathological sections of stomach and intestine were taken and the contents of motilin (MTL) and gastrin (GAS) were determined by enzyme-linked immunosorbent assay ( ELISA). Mifepristone combined with miso? prostol and ice water were used to establish the rat model of uterine bleeding caused by deficiency cold. Rats were randomly divid? ed into blank group, positive group and high, medium and low dose groups of decoction of processed ginger for 7 days. After the last administration,the uterus was sacrificed and pathological sections were taken to determine the four coagulation items and the daily blood loss. Results: All dosage groups of processed ginger can significantly improve the appearance of spleen-stomach deficiency cold bleeding model and uterine bleeding model rats, increase food intake and water intake, significantly improve four indicators of coagulation, shorten coagulation time. MTL and GAS content in spleen-stomach deficiency cold bleeding model rats increased significantly. Pathological sections showed that compared with model group, tissue damage was lighter and pathological changes were mild and shallow. The amount of bleeding decreased in deficiency cold uterine bleeding model rats. Conclusion: The pharmacologi cal action of processed ginger has the effect of warming channel for arresting bleeding.
