摘  要:目的:探讨白子菜总黄酮对脑缺血模型大鼠的影响,并探讨其可能的作用机制。方法:60只SPF级wistar大鼠随机分为假手术组、模型组、阳性对照组(尼莫地平片,20 mg·kg-1)、白子菜总黄酮大、小剂量组(200 g·kg-1、100 g·kg-1)。造模大鼠预防性给药14 d,末次给药2 h后,大鼠两侧颈总动脉用微动脉夹夹闭30 min,复制脑缺血模型。取大鼠脑组织,一半脑组织9倍生理盐水组织匀浆,酶免疫法检测大鼠脑组织匀浆液中白细胞介素-6(interleukin-6,IL-6)、白细胞介素-1β(interleukin-1β,IL-1β)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、诱导型一氧化氮合酶(inducible nitric oxide synthase,i Nos)、一氧化氮(nitric oxide,NO)含量。一半脑组织采用体积分数10%福尔马林固定液固定,切片,观察脑组织海马区病理情况。结果:与假手术组比较,模型组大鼠脑组织IL-6、IL-1β、TNF-α、i Nos、NO含量显著提高(P <0. 01);与模型组比较,白子菜总黄酮能降低大鼠脑组织IL-6、IL-1β、TNF-α、i Nos、NO含量(P <0. 01,P <0. 05),显著改善缺血所致脑组织海马区损伤。结论:白子菜总黄酮对脑缺血模型大鼠有较好的改善作用。Objective: To explore the effect of total flavonoids of Gynura divaricata on rat of cerebral ischemia models and to explore its possible mechanism. Methods: 60 Wistar male rats in grade SPF were randomly divided into a sham-operated group,a model group,a positive control group( Nimodipine pill,20 mg·kg-1),and a high and low dose of total flavonoids of Gynura divaricata( 200 g·kg-1,100 g·kg-1). After 14 days of prophylactic administration and 2 hours of last administration,both sides of common carotid artery were clamped with arterioles for 30 minutes to reproduce cerebral ischemia model. Rat brain tissues and half brain tissues were homogenized with 9 times normal saline. The levels of interleukin-6( IL-6),interleukin-1β( IL-1β),tumor necrosis factor-α( TNF-α),inducible nitric oxide synthase( i Nos),nitric oxide( NO) in rat brain homogenate were detected by enzyme immunoassay. Half of the brain tissues were fixed with 10% formalin and sliced to observe the pathological changes of hippocampus. Results: Compared with the sham-operated group,the levels of IL-6,IL-1β,TNF-α,i Nos and NO in brain tissue of model group were significantly increased( P < 0. 01);compared with model group,total flavonoids of Gynura divaricata can decrease the levels of IL-6,IL-1β,TNF-α,i Nos and NO in brain tissue of rats( P < 0. 01,P < 0. 05),and significantly improve hippocampal injury induced by ischemia. Conclusion: Total flavonoids of Gynura divaricata can improve cerebral ischemia animal model in rats.
