摘 要:目的:探讨中风解郁汤对卒中后抑郁(post-stroke depression,PSD)模型大鼠白细胞介素10(interleukin-10,IL-10)、IL-23与大鼠行为学的影响。方法:60只SD大鼠按照随机对照分组方法随机分为6组,分别为正常组、模型组、早期干预治疗组、进展期干预治疗组、早期对照组、进展期干预组,每组均为10只。应用改良的线栓法阻断SD大鼠的右侧大脑中动脉以制备梗死模型,再采用慢性不可预见的温和性应激及孤养法制备PSD大鼠模型,并在早期及进展期给予不同干预措施,早期界定于6 h以内,进展期界定于12 h^1周。治疗组给予中风解郁汤,对照组给予氟西汀胶囊,正常组、模型组不予任何处理,自由饮水、进食。14 d后,比较各组大鼠血清IL-10、IL-23水平,体质量,神经功能评分及行为学情况。结果:①神经评分方面:早期干预治疗组与进展期治疗组比较,差异有统计学意义(P=0.043<0.05)。②行为学方面:早期干预治疗组水平穿越格数与进展期治疗组比较,差异有统计学意义(P=0.02<0.05);早期干预治疗组站立次数与进展期治疗组比较,差异无统计学意义(P=0.161>0.05)。③炎症因子:早期干预治疗组IL-10与进展期治疗组比较,差异有统计学意义(P=0.000<0.01);早期对照组与进展期对照组比较,差异有统计学意义(P=0.004<0.01);早期干预治疗组IL-23与进展期干预治疗组比较,差异有统计学意义(P=0.005<0.01)。结论:中风解郁汤能改善PSD大鼠的神经功能评分及抑郁行为,可显著降低卒中后抑郁大鼠模型血清炎症因子的水平。Objective: To explore the effects of Zhongfeng Jieyu Decoction on interleukin-10 ( IL-10), IL-23 and behavior of rats with post-stroke depression ( PSD) model. Methods:60 SD rats were randomly divided into six groups:normal group,model group, early intervention group, advanced intervention group, early control group and advanced intervention group. Each group consisted of 10 rats. The right middle cerebral artery of SD rats was blocked by modified thread embolization to establish the infarction model. The PSD rat model was established by chronic unpredictable mild stress and solitary culture. Different interventions were given at the early and advanced stages, which were defined within 6 hours in the early stage and 12 hours to 1 week in the advanced stage. The treatment group was given Zhongfeng Jieyu Decoction, and the control group was given fluoxetine capsule. The normal group and the model group were not treated,and they were free to drink water and eat. After 14 days,serum IL-10,IL-23 levels,body mass, neurological function scores and behavioral status were compared between the groups. Results:(1)Neurological score: There was significant difference between early intervention group and advanced treatment group (P =0. 043 <0.05 ).②There was a significant difference in the number of horizontal crossings between the early intervention group and the advanced treatment group (P = 0. 02 <0. 05 );there was no significant difference in the number of standing between the early intervention group and the advanced treatment group (P=0. 161 >0. 05).③inflammatory factors:There was significant difference between the early intervention group and the advanced treatment group (P = 0. 000 < 0. 01 );there was significant difference between the early control group and the advanced control group (P =0. 004 <0. 01 );there was significant difference between the early intervention group and the advanced intervention group (P =0. 005 <0.01 ).
