益气汤含药血清诱导骨髓间充质干细胞干预大鼠骨性关节炎实验研究

摘　　要：目的:探讨益气汤含药血清诱导骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)对骨性关节炎大鼠的干预作用。方法:取SD大鼠骨髓,筛选、纯化并鉴定BMSCs,分别给予生理盐水与益气汤干预后获取血清。将实验动物分为对照组、生理盐水组、益气汤组。含药血清干预后观察大鼠毛色、食欲及活动情况等,同时测量大鼠体质量,治疗6周后取膝关节病理检查。结果:益气汤血清干预BMSCs电镜下观察细胞形态均一,边界清楚。生理盐水组膝关节炎程度退变明显。骨性关节炎模型大鼠运用益气汤含药血清干预BMSCs治疗后,原发病变缓解,关节组织滑膜病理检查均未见明显异常。结论:益气汤含药血清诱导下BMSCs能有效修复骨性关节炎大鼠软骨的退变和损伤。Objective: To explore the therapeutic effect of Yiqi Decoction drug-containing serum on bone marrow mesenchymal stem cells ( BMSCs) in rats with osteoarthritis. Methods: Bone marrow of SD rats was taken, BMSCs were screened,purified and identified ,and serum was obtained by normal saline and Yiqi decoction, respectively. Animal groups were model group, control group, saline group and Yiqi Decoction group. After treatment, the rats Appetite and activity were observed, and their body weight was measured. After 6 weeks of treatment, the knee joints were taken for pathological examination. Results: The cell morphology of BMSCs treated with Yiqi Decoction serum was uniform and the boundary was clear under electron microscope. The degeneration of knee joint arthritis was obvious in saline group. After BMSCs treatment with Yiqi Decoction, the primary lesion of osteoarthritis rat model was alleviated, and no obvious abnormality was found in synovial membrane pathological examination of joint tissue. Conclusion: BMSCs induced by Yiqi Decoction drug-containing serum can effectively repair cartilage degeneration and damage in rats with osteoarthritis.
