摘 要:目的探索回转模拟失重对RAW264.7巨噬细胞环状RNA(circRNA)表达谱的影响。方法RAW264.7细胞分为对照组(Con)和回转模拟失重组(Clino),Clino组以细胞回转器回转培养72h。采用Arraystar环状RNA芯片检测两组巨噬细胞circRNAs表达谱变化,筛选部分差异表达circRNAs,以qRT-PCR技术进行验证。结果与对照组相比,回转模拟失重组有60个circRNAs表达发生显著变化(差异倍数log2FC>1.5或<-1.5,P<0.05),其中上调34个,下调26个。qRT-PCR结果显示,circR003780,circR015248,circR015947及circR011728差异表达情况与芯片结果一致,差异具有统计学意义。结论回转模拟失重可改变RAW264.7巨噬细胞内circRNAs的表达模式,差异表达circRNAs可能通过调控巨噬细胞基本生理功能参与航天飞行导致的免疫功能改变。Objective To explore the effect of clinostat simulated weightlessness on the expression profiles of circular RNAs(circRNAs)in RAW264.7 macrophages.Methods RAW264.7 macrophages were divided into the control group(Con)and simulated weightlessness induced by clinostat group(Clino).The cells in Clino group were cultured in clinostat for 72 h.Arraystar circRNAs microarray was used to detect the changes of circRNAs expression profiles between Clino and Con groups,and quantitative real-time PCR(qRT-PCR)was applied to verify the expression of selected differentially expressed circRNAs.Results Compared with control group,there were 60 significant differentially expressed circRNAs in Clino group(Fold change log2 FC>1.5 or<-1.5,P<0.05),in which 34 circRNAs were upregulated and 26 down-regulated.qRT-PCR results showed that the expression of circR003780,circR015248,circR015947 and circR011728 were consistent with the results of circRNA microarray,the difference reached the statistical significance.Conclusion Simulated weightlessness induced by clinostat changed the expression profiles of circRNAs in RAW264.7 macrophages and differentially expressed circRNAs may be involved in the changes of immune functions induced by spaceflight through the regulation of basic physiological functions in macrophages.
