摘 要:为了探究亚硝酸盐胁迫对草鱼(Ctenopharyngodonidellus)肝脏泛素-蛋白酶体系统(ubiquitinproteasomesystem,UPS)的影响,将草鱼随机暴露于0、0.5、20mg/L的亚硝酸盐溶液中,在12、24、48、96h后采集肝脏样品,检测血液的生化指标,nrf2、hsp70和UPS相关基因的表达,泛素化蛋白的含量以及肝脏的组织学变化。结果显示:在亚硝酸盐胁迫12h,血清皮质醇含量呈现显著的剂量依赖式升高,且在24h和48h显著高于对照组。谷丙转氨酶和谷草转氨酶的含量在96h显著性降低。暴露12h时,20mg/L暴露组的nrf2、hsp70的表达量显著性高于对照组,ub、chip、psma2、psmc的表达水平和泛素化蛋白的含量在20mg/L亚硝酸盐胁迫24h均出现显著升高。在高浓度亚硝酸盐胁迫96h后,肝细胞出现明显的空泡化。研究表明,亚硝酸盐胁迫使草鱼产生了显著的应激反应,造成了肝组织损伤,使肝脏UPS活性上升。Ctenopharyngodon idellus were randomly exposed to three nitrite concentrations ( 0,0.5,and20.0 mg/L)for 96 h to evaluate the effect of nitrite stress on the ubiquitin-proteasome system ( UPS)in the liver.The expression of genes,the content of ubiquitinated protein,histological alterations of liver,and the levels of serum biochemical parameters were investigated at12,24,48 and 96 h after exposure,respectively.Compared to the control group,serum cortisol level was significantly elevated at12 h after exposure in a dose-dependent manner.Meanwhile,the significant rising in serum cortisol level was detected at24 and48 h.Serum levels of alanine aminotransferase and aspartate aminotransferase were significantly decreased at 96 h.The mRNA levels of nrf2 and hsp70 increased significantly at12 h after exposure (20.0 mg/L).Nitrite exposure of20.0 mg/L caused the significant elevations in mRNA expressions of ub,chip,psmc1,and psma2 as well as in ubiquitinated protein content at24 h.Hepatic vacuolization was observed in the liver of grass carp at 96 h after exposure to the concentration of nitrite (20.0 mg/L).These results suggested that the exposure of nitrite enhanced the activity of UPS as well as caused stress and histological damage of liver in C.idellus .
