摘 要:【目的】观察太子参试管苗继代培养过程中特殊的生物学性状表现,为太子参的应用开发研究提供理论依据。【方法】将太子参试管苗继代至芽扩繁培养基(MS+BA0.5mg/L)及生根培养基(1/2MS及1/2MS+NAA0.2mg/L),观察小苗生长过程中的生物学性状变化。【结果】在所测试的任何一种培养基上,培养60d后,试管苗均出现结参,并陆续出现开花、结籽现象。除块根,太子参长出丛根以及大量的须根,且不依赖于激素。随培养时间延长,试管苗茎节可以再生出大量气生根。【结论】太子参在试管内可以完成结参、开花、结籽等整个生命周期,且再生小苗大小适中,适用于制作离体植物微景观工艺品,用于科普及艺术观赏开发;试管内所结微块根,可为育种提供优质种源,也可作为直接提取太子参有效成分的原材料;太子参内源生长素比较旺盛,丛生根以及不定根的发生能力均较强,可用于器官培养途径——根培养途径生产次生物质的探索开发。【Objective】 In order to provide theoretical basis for the application and development of Pseudostellaria heterophylla (Miq.)Pax ex Pax et Hoffm, a common Chinese medicinal herb used for both medicine and food, some special biological characters were observed during subculture of the test-tube plantlets.【Methods】 The test-tube plantlets of P. heterophylla were subcultured to bud propagation medium (MS+BA 0.5 mg/L) and rooting medium (1/2MS and 1/2MS+NAA 0.2 mg/L), and then the changes of biological characters were observed.【Results】 After 60 days of culture on any tested medium, the plantlets produced microtuber root, bloom and seed. In addition to tuberous roots, P. heterophylla also grew tufted roots and a large number of fibrous roots without hormone. With the extension of culture time, a large number of aerial roots could be regenerated from the stem nodes of test-tube plantlets.【Conclusion】 P. heterophylla could complete the whole life cycle in vitro , including producing tuberous root, flower, and seed,etc. The plantlet size was moderate, and it was suitable for the production of isolated plant micro-landscape horticultural crafts which could be used for popular science and art viewing. The microtubule roots in vitro could provide high quality provenance for breeding and also be used as raw materials for extracting effective components of P. heterophylla directly. The endogenous auxin of P. heterophylla was exuberant, and the ability of tufted root and adventitious root formation was strong, which could be used to explore and develop secondary metabolite through root culture pathway.
