摘  要:目的探讨铁皮石斛的高效液相色谱(HPLC)各色谱峰与免疫力低下小鼠巨噬细胞吞噬功能的关系。方法从4个不同产地采集铁皮石斛,分别提取制成水、醇提取物,制备HPLC图谱。选取100只小鼠,随机分成正常组、模型组及8个样品组,每组10只。样品组给予不同产地铁皮石斛水(醇)提取物灌胃,正常组、模型组均灌胃蒸馏水。给药期间模型组及各样品组采用环磷酰胺法建立免疫低下小鼠模型。末次给药后,按0.1mL/10g尾静脉注射20%印度墨汁,计算廓清指数。应用偏最小二乘回归法分析廓清指数与指纹图谱间的谱效关系。结果建立了8个不同的铁皮石斛提取物HPLC指纹图谱,并获得30个共有特征色谱峰。模型组的廓清指数低于正常组(P<0.05),各铁皮石斛水提取物组廓清指数均高于模型组(均P<0.05)。色谱峰与铁皮石斛药效的相关性较强,可增强巨噬细胞吞噬功能的色谱峰为第4、6、8、10、11、14、15、17、18、21、23、27号。结论增强机体免疫功能的铁皮石斛有效成分主要为第4、6、8、10、11、14、15、17、18、21、23、27号色谱峰所代表的物质组分。Objective To investigate the relationship between each chromatographic peak of high performance liquid chromatography(HPLC) of Dendrobium officinale and macrophage phagocytosis in mice with hypoimmunity. Methods The water extract and ethanolic extract were made from Dendrobium officinale from 4 different regions,and then HPLC was used to produce spectrum.A hundred mice were selected and randomly divided into normal group,model group and 8 sample groups,with 10 mice in each group.Intragastric administration of water/ethanolic extracts of Dendrobium officinale from different regions was performed in the sample groups,and intragastric administration of distilled water in the normal group and model group.During administration,cytoxan-induced hypoimmunity mice model was established in the model group and each sample group.After the last administration,20% Indian ink(0.1 mL/10 g) was injected through the tail vein,and then the expurgation index was calculated.The partial least squares regression analysis was used to analyze the spectrum-effect relationship between expurgation index and fingerprint spectrum. Results The HPLC fingerprint spectrums of 8 different extracts from Dendrobium officinale were established,and 30 chromatographic peaks with shared characteristics were confirmed.The expurgation index of the model group was lower than that of the normal group( P <0.05), and the index of each Dendrobium officinale water extract group was higher than that of the model group(all P <0.05).The chromatographic peak was closely related to the efficacy of Dendrobium officinale ,and No.4,No.6,No.8,No.10, No.11 ,No.14,No.15,No.17,No.18,No.21,No.23 and No.27 chromatographic peaks exerted enhancement effect on macrophage phagocytosis. Conclusion The substances representing No.4,No.6,No.8,No.10,No.11,No.14, No.15,No.17,No.18,No.21,No.23 and No.27 chromatographic peaks are the major active components of Dendrobium officinale that enhance immune function of the body.
