摘  要:本试验通过对比健康与腹泻食蟹猴粪便菌群的差异,初步探讨腹泻对食蟹猴粪便菌群的影响。采集成年腹泻与健康食蟹猴粪样各6份,用细菌16Sr DNA通用引物扩增V3—V4 区,采用Illumina MiSeq测序平台对食蟹猴粪便微生物进行研究。结果表明,Alpha多样性指数分析,健康组与腹泻组的微生物多样性指数( Shannon、Simpson )差异不显著,群落丰富度指数( Chao、Ace )差异显著。PCA主成分分析,健康组与腹泻组粪便菌群具有明显的差异。通过对健康组与腹泻组物种菌群组成分析,运用MetaStat方法分析,与健康组相比,腹泻组的芽孢杆菌纲、乳酸杆菌目、乳酸杆菌属含量显著降低( P <0.05 ),腹泻组 Subdoligranulum 属显著升高( P <0.05 )。LEfSe ( LDA Effect Size )分析( LDA阈值为1 ),乳杆菌目,芽孢杆菌纲,乳杆菌科,乳酸杆菌属在健康组中显著富集,弯曲杆菌科,弯曲杆菌属在腹泻组显著富集。与健康食蟹猴相比,腹泻食蟹猴微生物区系中的一些菌群发生变化,菌群结构有差异但不显著( P >0.05 ),腹泻食蟹猴菌群结构有简单化的趋势。We investigated the influence of diarrhea on fecal microbiota of crab-eating macaque ( Macaca fascicularis ) by comparing the fecal microbiota of healthy and diarrheal crab-eating macaques.We collected fecal samples of 6 healthy and 6 diarrheal adult macaques,amplified the V3-V4 region using universal primer pairs of 16S rDNA for bacteria,and analyzed the amplicon using Illumina MiSeq sequencing platform.Alpha diversity index analysis showed that Shannon s diversity and Simpson s evenness indices were similar between healthy and diarrheal groups ( P >0.05 ),while chao and ace indices differed significantly between groups.Principal component Analysis ( PCA ) revealed significant difference of microbiota between groups.MetaStat analysis indicated that the abundance of Bacilli,Lactobacillales and Lactobacillus in the diarrhea group were significantly lower than in the healthy group ( P <0.05 ),and Subdoligranulum in the diarrhea group was significantly more abundant ( P <0.05 ).LEfSe analysis ( LDA=1 ) showed that Lactobacillales,Bacilli,Lactobacillaceae and Lactobacillus were significantly enriched in the healthy group,and Campylobacteraceae and Campylobacter were significantly enriched in the diarrheal group.The results prove that the fecal flora structure was similar between the two groups but the structure of gut flora tended toward simplification in the diarrheal group.
