摘  要:目的:评价介类中药牡蛎、石决明、瓦楞子煅制前后对肝阳上亢型高血压大鼠的降压作用,并探讨"介类潜阳"的药效组分及作用机制。方法:选用Wistar-Kyoto (WKY)大鼠为正常对照,自发性高血压大鼠(Spontaneously hypertensive rats,SHR)灌服附子汤5周复制肝阳上亢型高血压大鼠模型,分别以不同浓度的牡蛎、石决明、瓦楞子生、煅品水煎液给药,治疗2周后观察各组大鼠易激惹程度、眼结合膜充血程度,采用无创尾动脉测量大鼠收缩压(Systolic blood pressure,SP),采用酶联免疫测定法(ELISA)检测血浆去甲肾上腺素(Norepinephrine,NE)、肾上腺素(Epinephrine,E)、一氧化氮(Nitric oxide,NO)、血管紧张素Ⅱ(Angiotensin Ⅱ,AngⅡ)、醛固酮(Aldosterone,ALD)含量。结果:与WKY大鼠比较,模型组大鼠眼结合膜充血程度较重,易激惹程度多表现为Ⅲ级,饮水量增加,面部温度及SP均明显升高,血浆NE、E、AngⅡ、ALD含量增加,血浆NO含量降低(P<0.05,P<0.01)。与模型组比较,卡托普利组、牡蛎组、石决明组、瓦楞子组大鼠毛发紧凑有光泽,眼结合膜充血及易激惹程度减轻,饮水量减少,面部温度及SP降低,血浆NE、E、AngⅡ、ALD含量减少,血浆NO含量升高(P<0.05,P<0.01);煅牡蛎组、煅石决明组、煅瓦楞子组大鼠毛发蓬松程度及色泽,眼结合膜充血及易激惹程度,饮水量、面部温度、SP、血浆NE、E、AngⅡ、ALD、NO含量与模型组大鼠比较差异均无统计学意义(P>0.05)。结论:牡蛎、石决明生品有降低肝阳上亢型高血压大鼠血压及改善大鼠一般症状的作用,其作用机制可能与下调血浆NE、E、AngⅡ、ALD含量,上调血浆NO水平有关,且平肝潜阳作用优于瓦楞子生品;三者煅品无明显降压作用;三者煅制前后降压作用的差异性提示有机成分可能是"介类潜阳"药物的药效组分。Objective: To evaluate the antihypertensive effect of such testacean Chinese medicines as ostreae concha,haliotidis concha and arcae concha on rats with hypertension of liver-yang hyperactivity type before and after calcination,and discuss the composition and effectiveness of " meditating yang with testacean medicine" and its functional mechanism.Methods:Wistar-Kyoto(WKY) rats was selected as the normal control group. Spontaneously hypertensive rats(SHR) received Fuzi tang by gavage for 5 weeks so as to replicate the model of rats with hypertension of liver-yang hyperactivity type. They were medicated with the decoction of crude ostreae concha,haliotidis concha,arcae concha and their calcined products in different doses respectively. After 2 weeks of treatment,observed the degree of irritability and conjunctival congestion of rats in each group,assessed systolic blood pressure(SP) of rats via non-invasive measurement in the rat tail artery,and detected the contents of norepinephrine(NE),epinephrine(E),nitric oxide(NO),angiotensin Ⅱ(Ang Ⅱ) and aldosterone(ALD) in plasma via enzyme-linked immunosorbent assay(ELISA). Results:Comparing with WKY rats,the degree of conjunctival congestion of rats in the model group was higher, the degree of irritability was mainly manifested in grade Ⅲ, water consumption was increased,facial temperature and SP were both significantly increased,the contents of NE、E,Ang Ⅱ and ALD in plasma were increased,and the content of NO in plasma was decreased(P < 0.05,P < 0.01). Comparing with the model group,the hair of rats in the captopril group, the ostreae concha group, the haliotidis concha group and the arcae concha group was thick and glossy, the degree of irritability and conjunctival congestion was decreased, water consumption was reduced,facial temperature and SP were both decreased,the contents of NE、E,Ang Ⅱ and ALD in plasma were decreased,and the content of NO in plasma was increased(P < 0.05, P < 0.01);comparing with the model group, there were no significant differenc
