携CXCL12抗体靶向超声造影与不同时期卵巢癌移植瘤新生血管密度的相关性研究

摘　　要：目的应用携CXCL12抗体的靶向超声造影剂(MBt),探讨裸鼠卵巢癌移植瘤模型不同时期靶向超声造影参数与移植瘤MVD的相关性。方法移植瘤模型随机分为(10、20、30、40、50、60d)6组,每组裸鼠移植瘤行普通(MBc)及靶向超声造影(MBt)检查,获取超声造影时间-强度曲线(TIC)参数;CD31计数裸鼠瘤体新生血管密度(MVD),分析各组靶向造影参数与移植瘤MVD的相关性。结果成功制备靶向超声造影剂;MBt组峰值强度(PI)均大于MBc组,在10、20、30、40d组间差异有统计学意义,(P<0.05);MBt组达峰时间(TTP)均小于MBc组,各组间差异有统计学意义(P<0.05);6组MVD差异有统计学意义(P<0.05),MBt组PI值与MVD在20、30d组间呈正相关(r=0.673、0.697,P<0.05),MBt组TTP值与MVD在20、30d组间呈负相关(r=-0.671、-0.707,P<0.05)。结论携CXCL12抗体靶向超声造影参数PI、TTP与裸鼠卵巢癌移植瘤MVD在20、30d组时具有相关性,靶向超声造影一定程度上可实现在体评价肿瘤新生血管情况。Objective To apply cxcl12 Antibody targeting ultrasound contrast agent(MBt)Analyzing the correlation between the imaging characteristics of targeted contrast enhanced ultrasonography and microvessel density(MVD)in nude mice with ovarian cancer at different stages.Methods The transplanted tumor models were randomly divided into 6 groups(10,20,30,40,50,60 d);Targeted ultrasound contrast agent(MBt)was prepared.Normal(MBc)and targeted ultrasound contrast agent(MBt)were performed successively on each group of nude mice tumors;Obtaining parameters related to time intensity curve(TIC)of contrast-enhanced ultrasound,then CD31 were used to detect MVD.Correlation between target contrast parameters and MVD of transplanted tumor was analyzed.Results The targeted ultrasound contrast agent carrying CXCL12 antibody was prepared;The peak intensity(PI)of MBtgroup was greater than that of MBc group,(10,20,30,40 dgroup,P<0.05).The peak time(TTP)in group MBt was less than that in group MBc(P<0.05);There was a significant difference in MVD between the 6 groups(P<0.05),The PI of target imaging parameters was positively correlated with MVD at,20 and 30 days(r=0.673,0.697,P<0.05),and the TTP of target imaging parameters was negatively correlated with MVD at 20 and 30(r=-0.671,-0.707,P<0.05).Conclusions CXCL12 antibody targeted ultrasound contrast with nude mice ovarian cancer xenograft MVD in 20 d,30 dgroup correlation.Targeted contrast-enhanced ultrasound(CEUS)can evaluate neovascularization of ovarian cancer in vivo to a certain extent,and has certain value for early diagnosis and evaluation of ovarian cancer.
