摘  要:为了评估IFN-λ1作为免疫佐剂能否提高乙型脑炎病毒DNA疫苗的免疫效应,利用RT-PCR方法扩增猪源IFN-λ1基因,构建重组真核表达载体pVAX1-IFN-λ1,通过Western-blot和间接免疫荧光试验鉴定免疫效果。结果表明,转染pVAX1-IFN-λ1重组质粒的BHK21细胞能够正确表达IFN-λ1;将重组IFN-λ1与pVAX1-prMEDNA疫苗免疫小鼠后,与对照组pVAX1-prME相比,IFN-λ1可以诱导小鼠产生乙型脑炎病毒EⅢ蛋白特异性抗体水平、乙脑病毒特异性中和抗体效价和淋巴细胞增殖水平显著提高。结论,重组猪源IFN-λ1作为乙型脑炎病毒DNA疫苗的分子免疫佐剂能够在小鼠动物模型上提高乙型脑炎病毒DNA疫苗的免疫效应。To evaluate whether IFN-λ1 as immune-adjuvant can improve the immune effect of Japanese encephalitis virus DNA vaccine,the recombinant pVAX1-IFN-λ1 plasmid was constructed after the porcine IFN-λ1 gene was amplified by RT-PCR.The immune effects were idendificated by Western-blot and indirect immunofluorescence assay.The results indicated that IFN-λ1 was accurately expressed in BHK21 cells transfected with the pVAX1-IFN-λ1 plasmid.Groups of mice were immunized with pVAX1-prME DNA vaccine and recombinant IFN-λ1,compared with the immunized group of pVAX1-prME DNA vaccine,IFN-λ1 can significantly enhance the induced anti-EⅢprotein specific antibody level,JEV-neutralizing antibody titers and the level of lymphocyte proliferation.In conclusion,typeⅢinterferon PoIFN-λ1 could enhance the immune responses of DNA vaccine of JEV in the mice model.
