摘  要:目的探究lncRNA调控绝经后骨质疏松的途径及其机制。方法采用小鼠建立模型进行对比研究。适应环境一周之后,摘除模型组小鼠双侧卵巢,对照组小鼠保留卵巢。给予相同的粮食喝水,保证其正常生长发育。饲养8周后提取RNA并进行后续检测实验。结果模型组小鼠骨组织和骨髓间充质干细胞中lncRNA HOTAIR的表达显著升高,成骨相关基因显著降低,成脂基因显著升高,过表达lncRNA HOTAIR能够降低成骨相关基因的表达,上调成脂相关基因的表达;敲减lncRNA HOTAIR能够升高成骨相关基因的表达,降低成脂相关基因的表达。结论lncRNA可以作为生物标记物应用到绝经后骨质疏松患者的诊断和治疗当中,值得在临床上推广应用。Objective To explore the pathway and mechanism of lncRNA regulating postmenopausal osteoporosis.Methods The model was established in mice and was compared.One week after adapting to the environment,bilateral ovaries were removed in the model group,while ovaries were retained in the control group. Give the same food and water to ensure its normal growth and development.RNA was extracted after 8 weeks of feeding and subsequent detection experiments were carried out.Results The expression of lncRNA HOTAIR in bone tissue and bone marrow mesenchymal stem cells of model mice was significantly increased,the osteogenic related genes were significantly decreased,and the adipogenic genes were significantly increased.Overexpression of lncRNA HOTAIR could decrease the expression of osteogenic related genes and increase the expression of adipogenic related genes.Knocking down lncRNA HOTAIR could increase the expression of osteogenic related genes.The expression of related genes decreased the expression of adipogenic related genes.Conclusion lncRNA can be used as a biomarker in the diagnosis and treatment of postmenopausal osteoporosis,which is worthy of clinical application.
