摘 要:自2007年A型塞内卡病毒(SVA)在猪体内被发现以来,给全球养猪业造成了严重的经济损失,SVA感染的典型症状包括猪口鼻部囊泡的形成及破裂,以及蹄部冠状带水疱引起的跛行。用RT-PCR从河南某猪场具有水疱样病变的猪水疱皮或水疱液中检测到了SVA的特异性核酸片段,并通过IBRS-2细胞进行SVA的分离和传代,将分离得到的SVA命名为CH-HNXC,扩增和测定了VP1基因序列后分析发现,分离株CH-HNXC与之前报道的2017年河南和福建分离株位于较近的进化分支中,与加拿大和泰国分离株亲缘关系较远,表明SVA流行和进化中由相近时间点和较近地理位置带来高的同源性;分离株攻毒猪鼻吻部和蹄部可见水疱和溃烂、跛行等,表明本分离株能引起SVA所致的典型临床症状,为SVA在国内的流行提供了新的证据,为进一步研究SVA的致病机制、开发诊断试剂和疫苗奠定了基础。Since the discovery of Senecavirus A (SVA) in pigs in 2007,it has caused serious economic losses to the global pig industry.Typical symptoms caused by SVA in pigs include ruptured vesicles and erosions on the snout and lameness associated with broken vesicles along the coronary band.In this study,the specific fragment of SVA genome was detected from the vesicular skin and fluid of the pigs in a pig farm,Henan province.The SVA virus was isolated and passaged by IBRS-2 cells,and then the isolate was named CH-HNXC.As the result of VP1 gene sequence analysis showed,the isolate CH-HNXC has a closer relationship with the previously reported isolates from Henan and Fujian in 2017,and has a far relationship with isolates from Canada and Thailand,indicating the high homology caused by a similar time periods and closer geographical location in the process of evolution.In the pathogenicity experiment,vesicles on the snout and lameness associated with broken vesicles along the coronary band were observed in the challenged pigs,indicating that the isolate CH-HNXC has the characteristics of typical clinical symptoms caused by SVA.In conclusion,this study provided new evidence for the prevalence of SVA in China,and laid the foundation for further research on the pathogenic mechanism and the development of diagnostic reagent and vaccines of SVA.
