摘 要:目的:建立糖尿病性干眼动物模型,观察复明片改善糖尿病性干眼大鼠角膜功能和泪液炎症的疗效。方法:将30只实验级SD大鼠分为正常对照组、糖尿病性干眼组及复明片处理组,每组10只。复明片处理组大鼠在成功制作糖尿病性干眼模型后,给予复明片处理,观察三组大鼠的角膜荧光素评分、泪膜破裂时间、大鼠基础泪液分泌量及泪液细胞因子白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)和白细胞介素1β(IL-1β)的表达,并采用免疫组化法检测大鼠结膜上皮组织中转化生长因子β1(TGF-β1)的表达。结果:复明片处理组大鼠的泪膜破裂时间为(3.32±0.25)s,泪液分泌试验棉线湿润的平均长度为(3.99±0.98)mm,均明显低于正常对照组,但明显高于糖尿病性干眼组,组间比较差异均有统计学意义(P<0.05)。糖尿病性干眼组大鼠的角膜荧光素染色评分明显高于复明片处理组和正常对照组,差异均有统计学意义(P<0.05)。糖尿病性干眼组大鼠TGF-β1的表达水平最高,且阳性率明显高于复明片组,差异有统计学意义(P<0.05)。与正常对照组比较,复明片组和糖尿病性干眼组大鼠的泪液炎性细胞因子IL-6、TNF-a和IL-1β水平均明显升高,其中糖尿病性干眼组大鼠上述指标水平明显高于复明片组,差异均有统计学意义(P<0.05)。结论:复明片可以降低大鼠结膜上皮TGF-β1的表达,降低泪液炎性因子水平,进而改善干眼症状。OBJECTIVE: To establish the animal model of diabetic dry eye, and observe the effects of Fuming tablet on corneal function and tear inflammation in diabetic dry eye rats. METHODS: Thirty experimental SD rats were divided into the normal control group, the diabetic dry eye group and the Fuming tablets treatment group, with 10 rats in each group. After the successful preparation of the diabetic dry eye model, the rats in the Fuming tablets treatment group were given Fuming tablets. The corneal fluorescein score, tear breakup time, basic tear secretion and expression of tear cytokines IL-6, TNF-a and IL-1β were observed among the three groups. The expression of TGF-1 in conjunctival epithelium of rats was detected by immunohistochemistry. RESULTS: The tear breakup time of the rats in the Fuming Tablets treatment group was(3.32±0.25) s, and the average length of cotton tears in the tear secretion test was(3.99±0.98) mm, which was significantly lower than that of the normal control group, but significantly higher than that of the diabetic dry eye group,with statistically significant differences(P<0.05). The corneal fluorescein staining scores of the diabetic dry eye group were significantly higher than those of the Fuming tablets and the normal control group(P<0.05). The expression level of TGF-1 in the diabetic dry eye group was the highest, and the positive rate was significantly higher than that in the Fuming tablets group, with statistically significant difference(P<0.05). Compared with the normal control group, the levels of tear inflammatory cytokines IL-6, TNF-a and IL-1β in the Fuming tablets group and the diabetic dry eye group were significantly increased, and the diabetic dry eye group was significantly higher than the Fuming tablets group, with statistically significant differences(P<0.05). CONCLUSIONS: Fuming tablet can reduce the expression of TGF-1 in conjunctival epithelium of rats, reduce the level of inflammatory factors in tears, and thereby improve the symptoms of dry eyes.
