摘 要:目的观察滋阴降火方对围绝经期综合征(menopausal syndrome, MPS)阴虚内热型大鼠单胺类神经递质、雌激素水平的影响,探讨其对神经内分泌系统的干预作用。方法将大鼠按随机数字表法分为假手术组,模型对照组,雌激素片组,更年安胶囊组,滋阴降火方低、中、高剂量组,每组10只。除假手术组外,其余各组大鼠制备MPS阴虚内热型大鼠模型。滋阴降火方低、中、高剂量组分别灌胃8.46、42.3、84.6 g/kg滋阴降火方水煎液,雌激素片组灌胃戊酸雌二醇片0.45 mg/kg,更年安胶囊组灌胃3.10 g/kg更年安胶囊混悬液,假手术组和模型对照组灌胃等体积生理盐水,1次/d,连续30 d。末次给药后12 h,采用放射免疫检测法检测各组大鼠血清E2、FSH、黄体生成素(luteinizing hormone, LH)、泌乳素(prolactin, PRL)水平,计算子宫系数,采用HPLC-电化学检测器检测下丘脑中单胺类神经递质多巴胺(dopamine, DA)、去甲肾上腺素(norepinephrine, NE)、5-羟色胺(5-hydroxytryptamine, 5-HT)、5-羟吲哚乙酸(5-hydroxyindole acetic acid, 5-HIAA)水平。结果与模型组比较,滋阴降火方低、中、高剂量组大鼠血清FSH[(4.39±0.22)IU/L、(2.89±0.91)IU/L、(2.84±0.98)IU/L比(5.51±0.24)IU/L]、LH[(14.48±0.24)IU/L、(11.46±0.33)IU/L、(5.28±1.31)IU/L比(15.02±0.37)IU/L]水平降低(P<0.05),E2[(39.84±6.08)pmol/L、(48.65±6.77)pmol/L、(64.96±7.97)pmol/L比(33.16±4.62)pmol/L]水平升高(P<0.05或P<0.01),下丘脑DA[(231.28±17.29)ng/kg、(211.72±13.96)ng/kg、(197.96±13.96)ng/kg比(253.07±8.98)ng/kg]、5-HT[(321.59±15.79)ng/kg、(298.79±17.16)ng/kg、(211.25±13.35)ng/kg比(401.63±19.12)ng/kg]、5-HIAA[(559.42±16.57)ng/kg、(510.59±13.25)ng/kg、(377.85±14.83)ng/kg比(803.16±17.39)ng/kg]水平及5-HT/NE[(0.48±0.02)、(0.43±0.03)、(0.27±0.02)比(0.67±0.02)]、5-HIAA/5-HT[(1.74±0.09)、(1.71±0.10)、(1.80±0.17)比(2.00±0.10)]比值降低(P<0.05),NE[(663.34±9.81)ng/kg、(695.94±10.54)ng/kg、(790.76±16.35)ng/kg�Objective To observe the effect of Ziyin-Jianghuo formula in treatment of Yin deficiency generating intrinsic heat type of the levels of monoamine neurotransmitters and estrogens in rats, and investigate its intervention in the neuroendocrine system. Methods There were 7 groups, which were sham operation group, model control group, estrogen tablet group, Gengnian capsule group, Ziyin-Jianghuo formula low, medium and high dose groups. Castration was performed by castration (extraction of ovaries) plus hot traditional Chinese medicine. The rats in the treatment group were given the above drugs 24 hours after the last administration of the hot Chinese medicine, once a day for 30 consecutive days. The sham operation group and the model control group were given an equal volume of purified water by simultaneous intragastric administration. The serum estradiol (E2), luteinizing hormone (LH), follicle stimulating hormone (FSH), prolactin (PRL) levels,uterine coefficient and monoamine neurotransmitters dopamine (DA), norepinephrine (NE), serotonin (5-HT), 5-hydroxyindoleacetic acid (5-HIAA) content in the hypothalamus were detected after 12 hours of the last treatment. Results Compared with the model group, the levels of serum FSH (4.39 ± 0.22 IU/L, 2.89 ± 0.91 IU/L, 2.84 ± 0.98 IU/L vs. 5.51 ± 0.24 IU/L), LH (14.48 ± 0.24 IU/L, 11.46 ± 0.33 IU/L, 5.28 ± 1.31 IU/L vs. 15.02 ± 0.37 IU/L) in the low, middle and high doses of Ziyin-Jianghuo formula significantly decreased (P<0.05), the levels of serum E2 (39.84 ± 6.08 pmol/L, 48.65 ± 6.77 pmol/L, 64.96 ± 7.97 pmol/L vs. 33.16 ± 4.62 pmol/L) significantly increased (P<0.05). The content of DA, 5-HT, 5-HIAA in the hypothalamus and the 5-HT/NE (0.48 ± 0.02, 0.43 ± 0.03, 0.27 ± 0.02 vs. 0.67 ± 0.02), 5-HIAA/5-HT (1.74 ± 0.09, 1.71 ± 0.10, 1.80 ± 0.17 vs. 2.00 ± 0.10) in the low, middle and high doses of Ziyin-Jianghuo formula significantly decreased (P<0.05), the content of NE (663.34 ± 9.81 ng/kg, 695.94 ± 10.54 ng/kg, 790.76 ± 16.35 ng/kg vs. 602.95 ± 1
